AIM: This study focused on the relationship between the HPA axis function and the heat pain threshold in the orofacial region upon cold pressor (CP) stimulation. METHODS: Ten healthy male individuals participated in this study. CP stimulation was applied to each participant, and their peripheral blood was collected 5 min before, during and 5, 15, 30, 45, 60 min after receiving CP. In addition, 5 of those 10 participants were selected at random and they experienced a mock CP trial on different days. The heat pain thresholds on the facial skin about 10mm anterior to the right external auditory canal (trigeminal V2 region) in each subject were simultaneously recorded 5 min before and 5, 30, 60 min after CP stimulation. The blood pressure and heart rate were continuously monitored throughout the course of the CP and mock trials using an electric blood pressure meter. RESULTS: Significant increases in the plasma concentration of cortisol, beta-endorphin and ACTH were induced by CP stimulation, while no significant increases were observed under the mock trial conditions. The blood pressure and heart rate showed concomitant increases during CP stimulation. In addition, the heat pain threshold in the orofacial region significantly increased after receiving CP stimulation. These results suggest that CP stimulation activated the HPA axis thereby increasing the heat pain threshold in the orofacial region in healthy individuals. CONCLUSIONS: This observed pain threshold increase might be due to the activation of an endogenous opioid system, such as increase in the circulating beta-endorphin levels.

The aim of this study was to evaluate the effect of treating titanium (Ti) with polyphosphoric acid on the attachment and proliferation of human bone marrow derived mesenchymal stem cells (hBMSCs). Cleaned Ti disks were immersed into three different concentrations of polyphosphoric acid solution (0.1, 1, and 10 wt %) and 10 wt % orthophosphoric acid solution for 24 h at 37 C. Ti immersed in distilled water for 24 h at 37 C served as control. The level of polyphosphoric acid that interacted with the Ti surface was determined by measuring the surface P/Ti ratio (atom%/atom%) using X-ray photoelectron spectroscopy. Degrees of cell attachment (1, 3, 5 h after cell seed) and proliferation (l, 3, 5, and 7 days after cell seed) on each treated Ti disk were evaluated by MTS assay. The mean surface P/Ti ratios increased in a polyphosphoric acid concentration dependent manner. A significantly higher cell attachment was found on Ti treated with polyphosphoric acid in contrast to untreated Ti (control) for all three culture periods. MTS assay also revealed that cell proliferation levels significantly increased following a polyphosphoric acid dose dependency. Ti surface treatment with orthophosphoric acid did not influence the cell attachment and proliferation. It was concluded that polyphosphoric acid treatment of Ti enhanced the attachment and proliferation of hBMSCs. (c) 2007 Wiley Periodicals, Inc.

70歳女。金銀パラジウム合金を用いたレジン前装冠ブリッジを装着したが、上唇のヒリヒリ感、ブヨブヨ感を訴え、パノラマX線で残存歯に僅かな水平的な骨吸収が認めれた。上顎前歯部ブリッジに対する金属アレルギーが疑いパッチテストを施行し陽性であれば、金属製の歯冠修復物を除去し、ハイブリッドセラミックなどの材料に置換すると説明し了解を得た。パッチテストの結果、塩化コバルト、塩化パラジウムに陽性反応を認めた。上顎前歯部ブリッジ金属(パラジウム)に対するアレルギーと診断し、口腔内に存在する全ての金銀パラジウム修復物、補綴物の除去を行い、上顎前歯部ブリッジに対しては即時重合レジンを用いた暫間ブリッジを装着し、それ以外の除去部位にはハイブリッドセラミックによるジャケット冠やインレーを装着した。ハイブリッドセラミックで製作を予定したが歯根が破折したためチタン製インプラント補綴修復を提案し長期曝露試験を行いアレルギーがなく使用可能となった。

Cell attachment is a crucial step in tissue regeneration. In this study, human bone marrow stromal cells (hBMSCs) were isolated, and the effects of CCN2 on their attachment were examined. CCN2 significantly enhanced the hBMSC attachment, and this enhanced cell attachment was mainly regulated by the C-terminal module of CCN2. This enhancement was negated by the anti-integrin alpha(v)beta(3) antibody and p38 MAPK inhibitor, and phosphorylation of p38 MAPK was detected upon the enhanced cell attachment mediated by CCN2. We thus conclude that CCN2 enhances hBMSC attachment via integrin-p38 MAPK signal pathway. Enhanced hBMSC attachment on hydroxyapatite plates by CCN2 further indicated the utility of CCN2 in bone regeneration.

Muscle contraction accelerates IL-6 mRNA expression in the rat masseter muscle 国際誌

Objective: : This study was conducted to determine if interleukin-6 (IL-6) and interleukin-1 beta (IL-1 beta) mRNA expression increase in response to muscle contraction caused by repetitive electrical stimulation of the rat masseter muscle.
Methods: : Male Wistar rats weighing 140-160 g were divided randomly into the following three groups: electrical stimulation (ES) group (n = 21), carrageenan injection (CI) group (n = 24), and ES under dantrolene sodium (muscle relaxant) injection (ESDI) group (n = 7). ES or CI was done to the left masseter; and mock ES or mock Cl to the right. Muscle tissues on both sides were sampled for total RNA isolation. Real-time RT-PCR was performed, with the cyclophilin A (CypA) mRNA level in each sample as an internal control. Mean relative IL-6 (il-6/cypA) and IL-1 beta (il-1 beta/cypA) mRNA levels were compared between the experimental and mock-treated sides within each group.
Results: Mean IL-6/CypA levels in the ES- or Cl-treated muscle significantly increased, without any significant incremental change observed in either mock-treated muscle. Interestingly, the increase in the il-6/cypA level caused by the ES was suppressed by the injection of dantrolene sodium in the ESDI group. Furthermore, the mean il-1 beta/cypA level in the Cl-treated masseter also significantly increased without any significant incremental change observed in the mock-treated muscle. However, there was no significant difference in the mean il-1 beta/cypA levels in the masseter between the ES- and the mock-treated sides.
Conclusions: : These results show that IL-6 mRNA expression in the rat masseter muscle was accelerated by the CI or by repetitive muscle contraction induced by ES. Since the mRNA level of IL-1 beta, a well-known proinflammatory cytokine, was not altered by the contraction, the accelerated IL-6 mRNA expression elicited by the muscle contraction does not seem to be related to local inflammation. Since the mRNA level of IL-1beta, a well-known proinflammatory cytokine, was not altered by the contraction, the accelerated IL-6 mRNA expression elicited by the muscle contraction does not seem to be related to local inflammation.
