Dual role of cytosolic GSH in the ABA signaling pathway and plasma membrane ion channel regulation in guard cells of Vicia faba. International journal

Abscisic acid (ABA) induces stomatal closure in higher plants under drought stress. Glutathione (GSH) negatively regulates ABA-induced stomatal closure and reactive carbonyl species (RCS) play a role as signal mediators downstream of reactive oxygen species production in ABA signaling pathway in Arabidopsis thaliana. Activation of slow (S-type) anion channels and inhibition of inward-rectifying potassium ion (Kin+) channels in the plasma membrane are essential for ABA-induced stomatal closure. However, there is limited evidence regarding role of GSH in the activation of S-type anion channels and the inhibition of Kin+ channels. We used Vicia faba to clarify the regulation of these ion channels by GSH and RCS. Pretreatment of guard-cell protoplasts with the GSH-supplementing agent, glutathione monoethyl ester (GSHmee), suppressed the activation of S-type anion channels and the inactivation of Kin+ channels induced by ABA. The pretreatment with the RCS scavenger carnosine suppressed the activation of S-type anion channels and the inactivation of Kin+ channels by ABA. On patch clamping guard-cell protoplasts, the addition of GSH to the pipette (cytosolic) buffer decreased the S-type anion currents and increased the Kin+ currents. These results suggest that cytosolic GSH is involved in ABA-induced stomatal closure via negative regulation of ABA signaling and via direct regulation of ion channel activities in V. faba.

Traditional Chinese Zhenjiang aromatic vinegar (Kozu) contains Fraglide-1 (FG1), a bioactive lactone with demonstrated peroxisome proliferator-activated receptor gamma (PPARγ) agonist and antioxidant activities. This study explored FG1's novel ability to antagonize the aryl hydrocarbon receptor (AhR) signaling pathway, which regulates artemin expression and contributes to itching and inflammation in atopic dermatitis. Through molecular docking simulations and cell-based assays in human keratinocytes, we demonstrated FG1's potent antagonistic activity against AhR signaling. FG1 effectively suppressed FICZ-induced inflammatory responses, including artemin expression, with potency (half maximal inhibitory concentration, IC50 = 5.1 μM) comparable to the synthetic antagonist StemRegenin 1 (SR1) while demonstrating a superior safety profile (median lethal concentration, LC50 > 100 μM vs. 27.5 μM for SR1). These findings expand our understanding of bioactive compounds from traditional fermented foods and their regulatory effects on AhR signaling, providing a foundation for future studies on FG1's role in modulating skin inflammation.

Myrosinase, referred to as thioglucoside glucohydrolase (TGG), plays a crucial role in plant physiology through catalyzing the hydrolysis of glucosinolates into bioactive isothiocyanates. In Arabidopsis thaliana, the myrosinases TGG1 and TGG2 are essential for abscisic acid- and methyl jasmonate-induced stomata closure. Allyl isothiocyanate (AITC), one of myrosinase products, triggers stomatal closure in A. thaliana. We investigated stomatal responses to AITC to clarify the role of TGG1 and TGG2 in Arabidopsis guard-cell signaling. Allyl isothiocyanate at 50 μM and 100 μM induced stomatal closure in the tgg1 and tgg2 single mutants but not in the tgg1 tgg2 double mutant. Furthermore, AITC at 50 μM induced the production of reactive oxygen species and nitric oxide, cytosolic alkalization, and oscillations in cytosolic free calcium concentration in guard cells of both wild-type and mutant plants. These findings suggest that TGG1 and TGG2 are involved in AITC signaling pathway through interaction with signal component(s) downstream of these signaling events, which is not accompanied by hydrolysis of glucosinolates because of the difference in subcellular localization between enzymes (myrosinases) and substrates (glucosinolates).

An elicitor, chitosan (CHT), induces stomatal closure in plants, which is accompanied by salicylhydroxamic acid (SHAM)-sensitive peroxidases-mediated reactive oxygen species (ROS) production in guard cells. Reactive carbonyl species (RCS) function downstream of ROS in abscisic acid (ABA) and methyl jasmonate (MeJA) signalling in guard cells. However, the involvement of RCS in CHT-induced stomatal closure is still unknown. In this study, we used transgenic tobacco (Nicotiana tabacum) plants overexpressing Arabidopsis thaliana 2-alkenal reductase (AER-OE tobacco) and Arabidopsis wild-type (WT) plants to investigate whether RCS is involved in CHT-induced stomatal closure. Chitosan-induced stomatal closure was inhibited in the tobacco AER-OE plants. In the WT tobacco and Arabidopsis plants, CHT-induced stomatal closure was inhibited by RCS scavengers, carnosine and pyridoxamine. Chitosan significantly increased RCS production in the WT tobacco and Arabidopsis, but in the tobacco AER-OE plants, chitosan did not increase significantly RCS accumulation. Moreover, neither the application of RCS scavengers to both WT plants nor scavenging RCS by AER-OE affected the CHT-induced ROS accumulation. However, treatment with a peroxidase inhibitor, SHAM, significantly inhibited CHT-induced RCS accumulation in WT tobacco and Arabidopsis plants. Taken together, these results suggest that RCS acts downstream of ROS production in CHT signalling in guard cells of A. thaliana and N. tabacum.

GUARD CELL HYDROGEN PEROXIDE-RESISTANT1 (GHR1), a leucine-rich repeat receptor-like kinase, is involved in abscisic acid (ABA)-induced stomatal closure. We investigated the role of GHR1 in reactive oxygen species (ROS) signaling for ABA-induced stomatal closure. ABA induced ROS production in wild type (WT) and the ghr1 of Arabidopsis thaliana. Hydrogen peroxide induced stomatal closure, accompanying the generation of acrolein in guard cells. The reactive carbonyl species (RCS) scavengers inhibited the ABA- and H2O2-induced stomatal closure in WT. In the ghr1, H2O2 failed to induce acrolein production and stomatal closure while RCS induced stomatal closure. Thus, GHR1 functions downstream of ROS and is required for the generation of RCS in guard-cell ABA signaling. In the ghr1, Ca2+ induced stomatal closure but RCS did not activate ICa channels. The GHR1 may be also involved in a Ca2+-independent pathway for ABA-induced stomatal closure.

Osmolytes proline (Pro) and glycine betaine (GB) have been reported to impart tolerance against salinity stress in many plants. However, there is no report available on the combined application of Pro and GB to mitigate salinity-induced growth inhibition in maize. Consequently, the goal of the current study is to assess Pro's and GB's potential as priming and exogenous agents in maize under salt stress. Therefore, the present study was conducted using a petri dish and hydroponic pot experiment to evaluate the morpho-physiological and biochemical characteristics of maize plants subjected to salt stress, with the addition of Pro and GB. The seeds of maize were germinated under 20 mM of each Pro or GB, with or without 120 mM salinity stress. The findings demonstrated that salt stress lessened the final germination percentage (FGP) (52.9 %), photosynthetic pigments (40.0 %), relative water content (RWC) (20.2 %), stomatal size (59.3 %), and leaf and root K+/Na+ ratios of maize seedlings compared to control. In addition, compared to the control, higher levels of malondialdehyde (MDA) (24.2 %) and hydrogen peroxide (H2O2) (25.7 %) were observed, whereas lower levels of catalase (CAT) (43.8 %) and peroxidase (POX) (30.4 %) were noted. The priming with Pro, GB, Pro+GB significantly increased FGP, germination index (GI), shoot-root biomass, seed vigor index (SVI), and reduced mean germination time (MGT) under salt stress. Foliar application of Pro, GB, or Pro+GB resulted in a significant increment in chlorophyll contents, RWC, K+ absorption, and stomatal size under salt stress. Furthermore, supplementing with Pro, GB, or Pro+GB reduced the accumulation of H2O2 (24.0, 23.3, and 31.1 %, respectively) and MDA (22.8, 17.2, and 32.1 %, respectively) caused by salt and augmented the levels of CAT (33.3, 22.8, and 45.2 %, respectively) and POX (36.4, 23.5, and 47.2 %, respectively) in the leaves. Taken together, the current study's findings indicate that combining Pro and GB is one of the most effective techniques for improving the salinity-tolerant seed germination and seedling traits of maize plants. Consequently, this study recommends that Pro and GB can be used as seed priming and exogenous agents to help maize grow faster in salt-stress situations.

Chilling is one of the major abiotic stresses for plants, especially for rice cultivation. Many essential metabolic processes for growth and development are temperature-dependent. In that case, reducing the negative effects of cold stress using exogenous chemicals is a possible option. Therefore, the current study examined the effects of pre-sowing seed treatment with different chemicals, viz. hydrogen peroxide (H2O2), salicylic acid (SA), calcium chloride (CaCl2), thiourea (TU), and citric acid (CA) on the germination of rice seeds (cv. BRRI dhan28) under chilling environments. Rice seeds were soaked in distilled water (control), 10 mM CA, 2 mM SA, 10 mM CaCl2, 10 mM H2 O2, and 10 mM TU solutions for 24 h. After that, seeds were exposed to chilling stress by incubating at 4 ± 1°C for 8 h, followed by at 25 ± 2°C for 16 h for 7 days. Exposure to chilling stress significantly reduced the final germination percent (13.6%), germination rate index (36.0%), coefficient of the velocity (25.0%), shoot fresh weight (44.4%), and root fresh weight (60.5%). Moreover, chilling induced oxidative damage and reduced the activity of antioxidant enzymes (catalase and ascorbate peroxidase). In contrast, treatments with H2O2, SA, CaCl2, TU, and CA considerably enhanced germination indices and seedling growth compared to chilling stress conditions. The study showed that priming with H2 O2, SA, CaCl2, TU, and CA significantly boosted antioxidant enzyme activities and reduced MDA and H2O2 contents in chilling-stressed rice plants, indicating less oxidative stress and improved tolerance. Principal component analysis showed that among these priming agents, H2O2, SA, and CA are most effective in chilling stress mitigation. Therefore, using seed-treating chemicals to combat the effect of chilling stress can help rice seedlings grow better in the winter season.

Plants often face high salinity as a significant environmental challenge with roots being the first responders to this stress. Maintaining K+/Na+ ratio within plant cells is crucial for survival, as the intracellular K+ level decreases and the intracellular Na+ level increases under saline conditions. However, knowledge about the molecular regulatory mechanisms of K+ loss in response to salt stress through outward-rectifying K+ channels in plants is largely unknown. In this study, we found that the Arabidopsis double mutant gorkskor, in which the GORK and SKOR genes are disrupted, showed an improved primary root growth under salt stress compared to wild-type (WT) and the gork and skor single-mutant plants. No significant differences in the sensitivity to mannitol stress between the WT and gorkskor mutant were observed. Accumulation of ROS induced by salt stress was reduced in the gorkskor roots. The gorkskor mutant seedlings had significantly higher K+ content, lower Na+ content, and a greater resultant K+/Na+ ratio than the WT under salt stress. Moreover, salt-stress-induced elevation of cytosolic free Ca2+ concentration was reduced in the gorkskor roots. Taken together, these results suggest that Arabidopsis Shaker-type outward-rectifying K+ channels GORK and SKOR may redundantly function in regulation of primary root growth under salt stress and are involved in not only the late-stage response (e.g. K+ leakage) but also the early response including ROS production and [Ca2+]cyt elevation.

Protective effect of quercetin against acetaldehyde was evaluated using the cultured hepatocyte models with aldehyde dehydrogenase (ALDH) isozyme deficiency (aldh2-kd and aldh1a1-kd). The quercetin-induced cytoprotection against acetaldehyde in the ALDH1A1-deficient mutant (aldh1a1-kd) was weaker than that in the wild type. Furthermore, quercetin did not enhance the ALDH activity in aldh1a1-kd cells, suggesting that ALDH1A1 is involved in quercetin-induced cytoprotection.

Chitosan (CHT) is a deacylated derivative of chitin and improves growth and yield performance, activates defensive genes, and also induces stomatal closure in plants. Glutathione (GSH) has significant functions in the growth, development, defense systems, signaling, and gene expression. GSH negatively regulates abscisic acid-, methyl jasmonate-, and salicylic acid-induced stomatal closure. However, the negative regulation by GSH of CHT-induced stomatal closure is still unknown. Regulation of CHT-induced stomatal closure by GSH in guard cells was investigated using two GSH-deficient mutants, cad2-1 and chlorina 1-1 (ch1-1), and a GSH-decreasing chemical, 1-chloro-2,4-dinitrobenzene (CDNB). The cad2-1 and ch1-1 mutations and CDNB treatment enhanced CHT-induced stomatal closure. Treatment with glutathione monoethyl ester restored the GSH level in the guard cells of cad2-1 and ch1-1 and complemented the stomatal phenotype of the mutants. These results indicate that GSH negatively regulates CHT-induced stomatal closure in Arabidopsis thaliana.

It is still unclear whether or how quercetin influences the toxic events induced by acetaldehyde in hepatocytes, though quercetin has been reported to mitigate alcohol-induced mouse liver injury. In this study, we evaluated the modulating effect of quercetin on the cytotoxicity induced by acetaldehyde in mouse hepatoma Hepa1c1c7 cells, the frequently used cellular hepatocyte model. The pretreatment with quercetin significantly inhibited the cytotoxicity induced by acetaldehyde. The treatment with quercetin itself had an ability to enhance the total ALDH activity, as well as the ALDH1A1 and ALDH3A1 gene expressions. The acetaldehyde treatment significantly enhanced the intracellular reactive oxygen species (ROS) level, whereas the quercetin pretreatment dose-dependently inhibited it. Accordingly, the treatment with quercetin itself significantly up-regulated the representative intracellular antioxidant-related gene expressions, including heme oxygenase-1 (HO-1), glutamate-cysteine ligase, catalytic subunit (GCLC), and cystine/glutamate exchanger (xCT), that coincided with the enhancement of the total intracellular glutathione (GSH) level. Tin protoporphyrin IX (SNPP), a typical HO-1 inhibitor, restored the quercetin-induced reduction in the intracellular ROS level, whereas buthionine sulphoximine, a representative GSH biosynthesis inhibitor, did not. SNPP also cancelled the quercetin-induced cytoprotection against acetaldehyde. These results suggest that the low-molecular-weight antioxidants produced by the HO-1 enzymatic reaction are mainly attributable to quercetin-induced cytoprotection.

Wheat is affected by various biotic and abiotic stresses, especially salinity, which reduces the growth and yield drastically. High salinity is a major constraint for wheat productivity in many countries, including Bangladesh. With this view, here, an experiment was conducted to observe genotypic differences in physiological, ion accumulation, agro-morphological and yield performance of wheat against different levels of salinity. Experimental variables consisted of five salt tolerant genotypes (G 20–2–2, G 20–1–2, G 13–2, G 22–2, G 9–2), one susceptible genotype (G 24–2) and one standard check variety (BARI ghom 25), which assigned to four levels of salinity with electrical conductivities control (0.3), 4, 8 and 12 dS m−1. Irrespective of genotypes, salinity stress significantly decreased the yield and yield attributes. Results of analyses based on salt tolerance indices of plant growth related and yield contributing parameters, ionic balance (Na+, K+ and Na+ /K+ ratio), and stress indicators such as chlorophyll content, photosynthetic rate (Pn), stomatal conductance (gs) and transpiration rate (Tr) revealed genotypes G 20–2–2, G 13–2 and G 20–1–2 as salt tolerant, genotype G 9–2 as moderately salt-tolerant and G 24–2 and G 22–2 as salt-sensitive genotypes. Additionally, lower accumulations of hydrogen peroxide and malondialdehyde, and higher activities of antioxidant enzymes in the salt-tolerant genotypes G 20–2–2 and G 13–2 than in the salt-sensitive genotype G 24–2 indicated reduced oxidative damage in genotypes G 20–2–2 and G 13–2 relative to that in genotype G 24–2. Collectively, our findings suggest that the optimum growth and yield of salt tolerant genotypes are associated with decreased Na+/K+ ratio, increased antioxidant enzymes activity and reduced oxidative stress.

Given the adverse impacts of heavy metals on plant development and physiological processes, the present research investigated the protective role of indole-3-acetic acid (IAA) and gibberellic acid (GA3) against cadmium (Cd)-induced injury in chickpea seedlings. Therefore, seeds germinated for 6 days in a medium containing 200 μM Cd alone or combined with 10 μM GA3 or 10 μM IAA. Both GA3 and IAA mitigated Cd-imposed growth delays in roots and shoots (80% and 50% increase in root and shoot length, respectively). This beneficial effect was accompanied by a significant reduction in Cd2+ accumulation in both roots (74% for IAA and 38% for GA3) and shoots (68% and 35%, respectively). Furthermore, these phytohormones restored the cellular redox state by reducing the activity of NADPH oxidase and downregulating the transcription level of RbohF and RbohD genes. Likewise, hydrogen peroxide contents were reduced by GA3 and IAA supply. Additionally, GA3 and IAA countered the Cd-induced reduction in total phenols, flavonoids, and reducing sugars in both roots and shoots. The exogenous effectors enhanced the activities of catalase, ascorbate peroxidase, and thioredoxin, as well as the corresponding gene expressions. Interestingly, adding GA3 and IAA to the Cd-contaminated germination media corrected the level of calcium (Ca2+) ion within seedling tissues. This effect coincided with the upregulation of key genes associated with stress sensing and signal transduction, including auxin-binding protein (ABP19a), mitogen-activated protein kinase (MAPK2), calcium-dependent protein kinase (CDPK1), and calmodulin (CaM). Overall, the current results suggest that GA3 and IAA sustain the Ca2+ signaling pathway, resulting in metal phytotoxicity relief. Amendment of agricultural soils contaminated with heavy metals with GA3 or IAA could represent an effective practice to improve crop yield.

Chitosan Enhances Drought Tolerance in Maize (<i>Zea may</i>s L.) by Promoting Growth and Chlorophyll Content While Reducing Hydrogen Peroxide Levels Reviewed International coauthorship International journal

Drought is one of the major constraints for maize (<i>Zea mays</i> L.) cultivation at the global level, as maize is sensitive to drought. To elucidate the impact of chitosan (CS) on improving drought tolerance in maize, the morphological and biochemical features of a drought-stressed maize variety (CAVARI 3696) at the germination and seedling stages were analyzed. The experiments performed at both stages were comprised of four different treatments in triplicate, viz., control (C), drought (D), drought with 50 ppm chitosan (D+50 CS) and 50 ppm chitosan only (50 CS). At the germination stage, drought stress significantly declined seed germination percentage and the growth and biomass of seedlings, while CS supplementation under drought enhanced germination and growth significantly. In the seedling stage, drought-stressed maize plants exhibited a significant reduction in chlorophyll contents (both Chl a and Chl b) but elevation in H2O2 buildup and lipid peroxidation measured as malondialdehyde (MDA) level. Drought stress also enhanced proline accumulation and improved catalase (CAT), peroxidase (POD) and ascorbate peroxidase (APX) enzyme activities in maize plants. However, chitosan raised chlorophyll levels and suppressed H₂O₂, MDA, and proline content in maize plants under drought stress. Moreover, CS supplementation reduced the activities of the antioxidant enzymes under drought. These results suggest that chitosan can be used to improve the growth performance of maize under drought through enhancement in defense responses against the buildup of reactive oxygen species and associated oxidative injury.

The aim of this study is to investigate the modulating effect of coexisting food components on the absorption and metabolism of quercetin and blood plasma antioxidant potentials. The combination of quercetin with α-tocopherol (αT), cellulose, or a commercially available vegetable beverage containing αT and dietary fiber was orally administered to mice. Compared to the single administration of quercetin aglycone, the coadministration of αT with quercetin significantly increased the plasma quercetin concentration at 0.5 h, whereas the combination of quercetin and cellulose decreased it. Interestingly, the administration of quercetin mixed with the vegetable beverage showed no significant change in the quercetin concentration in the mice plasma. The treatment of the cells with the blood plasma after the coadministration of αT with quercetin significantly upregulated the gene expression of the antioxidant enzyme (heme oxygenase-1), whereas the quercetin and cellulose combination did not. In the plasma of the quercetin-administered mice, eight types of quercetin metabolites were detected, and their quantities were affected by the combination with αT. The potentials of the heme oxygenase-1 gene expression by these metabolites were very limited, although several metabolites showed radical scavenging activities comparable to aglycone in the in vitro assays. These results suggested that the combination of αT potentiates the quercetin absorption and metabolism and thus the plasma antioxidant potentials, at least in part, by the quantitative changes in the quercetin metabolites.

Temperature sensing is critical for the survival of living organisms.1,2 Thermosensitive transient receptor-potential (TRP) cation channels function as thermosensors in mammals.2,3,4,5,6 In contrast to animals, land plants lack TRP genes.7,8,9 Previous patch-clamp studies in plant cells suggested the presence of ion channels whose activities are related to temperature, implying the presence of TRP-like channels.10,11,12,13,14 However, the molecular entities of such temperature-sensitive ion channels were still unknown in land plants. In this study, we observed that the unique rainfall-induced leaf-folding movement of the legume tree Samanea saman15 was temperature-sensitive by using a rainfall-mimicking assay. Chilling-induced leaf folding in S. saman was shown to be related to the swelling of the motor cells16,17 at the base of the leaflet. This swelling suggested involvement of temperature-sensitive inactivation of K+ currents, independent of fluctuations in ion channel gene expression in motor cells. These findings led us to examine the temperature sensitivity of an outward-rectifying K+ channel, SPORK2, which was reported as an ion channel responsible for the nyctinastic (circadian-rhythmic) leaf movement of S. saman.18 We also discovered that SPORK2 exhibits temperature-sensitive K+ transport activity in the Xenopus oocyte expression system. Using chimeric channels, we showed that two domains of SPORK2 regulated the temperature sensitivity. Furthermore, heterologously expressed SPORK2 in Arabidopsis guard cells induced temperature-dependent stomatal closure. Therefore, SPORK2 is an ion channel in land plants with temperature-sensitive ion-transport activity that functions similarly to mammalian TRP channels. Our current findings advance the molecular understanding of temperature-sensing mechanisms in plants.

Dihydroxyacetone (DHA) occurs in wide-ranging organisms, including plants, and can undergo spontaneous conversion to methylglyoxal (MG). While the toxicity of MG to plants is well-known, the toxicity of DHA to plants remains to be elucidated. We investigated the effects of DHA and MG on Arabidopsis. Exogenous DHA at up to 10 mm did not affect the radicle emergence, the expansion of green cotyledons, the seedling growth, or the activity of glyoxalase II, while DHA at 10 mm inhibited the root elongation and increased the activity of glyoxalase I. Exogenous MG at 1.0 mm inhibited these physiological responses and increased both activities. Dihydroxyacetone at 10 mm increased the MG content in the roots. These results indicate that DHA is not so toxic as MG in Arabidopsis seeds and seedlings and suggest that the toxic effect of DHA at high concentrations is attributed to MG accumulation by the conversion to MG.
