Immunohistochemical study on leukocyte adhesion molecules expressed on lymphatic endothelium

Leukocyte adhesion molecules expressed on the lymphatic endothelium in human small intestine and submandibular lymph node were studied immunohistochemically. Lymphatic capillaries in the lamina propria, mucosal muscle layer, and submucosal connective tissue of the intestine and in the capsule of the lymph node showed strong expression of platelet-endothelial cell adhesion molecule-1 (PECAM-1). A few lymphatic capillaries that weakly expressed intercellular adhesion molecule-1 (ICAM-1) were found in the capsule of the lymph node but in the small intestine, no lymphatic capillaries expressed detectable amounts of ICAM-1. Lymphatic capillaries also did not express detectable amounts of endothelial cell-selectin in the small intestine and lymph node. When lymphocytes migrate from tissue into lymphatic capillaries, multiple adhesion molecules may not be required for the migration. PECAM-1, however, may contribute to adherence of lymphocytes to lymphatic endothelium and the expression of adhesion molecules on lymphatic endothelium may be different between tissues. Previously, lymphatic endothelium of human tissue has been shown to express only platelet-endothelial cell adhesion molecule-1 (PECAM-1). In this study we examined the expression of immunoglobulin superfamily members on the lymphatic endothelium of human small intestine while in the presence of inflammatory cytokines. Lymphatic vessels were identified by using a cocktail of IgGs for desmoplakin I and II while the presence of inflammatory cytokines was determined by the expression of major histocompatibility complex (MHC) class II in the venules. As a result, lymphatic vessels in the tissue with venules expressing MHC class II expressed PECAM-1, intercellular adhesion molecule (ICAM)-1, ICAM-3, and vascular cell adhesion molecule-1 (VCAM-1). The expression of ICAM-3 and VCAM-1 was significantly stronger in lymphatic vessels than in blood vessels. The results suggest that inflamed lymphatic endothelium may allow more lymphocyte subpopulations to adhere to the endothelium than non-inflamed lymphatic endothelium, due to the expression of multiple adhesion molecules playing a role. The existence of lymphatic vessels in dental pulp has been a matter of continuing controversy because of the difficulty of discriminating them in ordinary stained tissue sections. Recently, we have succeeded in establishing a new identification method for lymphatic vessels in human frozen sections by using a commercial monoclonal antibody specific for the human thoracic duct and anti-human laminin antiserum. The present study aimed to examine the lymphatic vessels in human dental pulp using the new immunostaining method, and compared the results with those in human small intestine. The study clearly demonstrated the distribution of lymphatic vessels in human dental pulp. Large lymphatic vessels are located in the central part of the pulp and there are small lymphatic vessels in the periphery of the pulp. This suggests that lymphatic drainage of the human dental pulp starts from the periphery of the pulp and collects in the central part of the pulp. A notable difference between the small intestine and dental pulp was found in the immunoreactivity of lymphatic vessels to anti-human laminin antiserum. In small intestine, immunoreactivity was significantly weaker than that of the blood vessels, whereas in dental pulp, that of lymphatic vessels was almost the same as blood vessels, except for some lymphatic vessels showing very weak reactivity. These findings suggest that the development of the basement membrane in both the lymphatic and blood vessels of human dental pulp is not as marked as in other tissue.

The expression of adhesion molecules on the lymphatic endothelium of human small intestine and submandibular lymph node was studied immunohistochemically with the antibodies for selectin family and Ig superfamily members. In both small intestine and submandibular lymph node, lymphatic endothelium did not express intercellular adhesion molecule-1 and endothelial cell-selectin but expressed platelet-endothelial cell adhesion molecule-1 (PECAM-1), Though lymphatic vessels may not have a positive function in leukocyte rolling and adhesion, lymphatic endothelium may interact with leukocytes, with PECAM-1 playing a role.

The expression of adhesion molecules on the vascular endothelium of healthy and inflamed human dental pulp was studied immunohistochemically using antibodies for selectin family and immunoglobulin superfamily members. In healthy dental pulp, some vessels reacted very weakly with major histocompatibility complex (MHC) class II, and E-and P-selectin, and many vessels reacted with platelet-endothelial cell adhesion molecule-1 (PECAM-1), but no vessels reacted with intercellular adhesion molecule-1 (ICAM-1), ICAM-3 and vascular cell adhesion molecule-1 (VCAM-1), in inflamed pulp, a large number of vessels reacted strongly with MHC class II, E-and P-selectin, PECAM-1, ICAM-1, ICAM-3, and VCAM-1. This indicates that the vascular endothelium of the inflamed human dental pulp expresses diverse adhesion molecules for leukocyte emigration from the blood stream into tissue. Dental pulp is surrounded by dentin and isolated from other tissue, like an in vitro system, so dental pulp is an attractive model for studying the roles of adhesion molecules on the vascular endothelium in transendothelial migration of leukocytes.

Light microscopic identification of lymphatic vessels of the human small intestine was studied immuno-histochemically with a monoclonal antibody specific for human thoracic duct (mAb-D) and rabbit anti-human laminin antibody (anti-L). Two types of vessels were observed with anti-L; one strongly reacted with regularly outlined vessels and the other weakly reacted with irregularly outlined vessels and thin walls. Immunoreactivity to mAb-D was strong in the weakly reacting vessels, and the strongly reacting vessels did not react to mAb-D. Immunoreactivity of lymphatic vessels to anti-L has been reported to be weaker than that of the blood vessels; only irregularly outlined vessels which were weakly immunostained with anti-L reacted to mAb-D and no cross-reaction to mAb-D was observed in the regularly outlined vessels. This indicates that the vessels strongly reacting with anti-L are blood vessels and the vessels immunostained with mAb-D must be lymphatics. The study shows that it is possible to identify lymphatic vessels light microscopically by mAb-D. This method will be useful in studying the fine distribution of lymphatic vessels in normal tissue and also in pathological tissue such as metastasis of malignant tumors. (C) 1997 Academic Press.

Human immunoglobulin G Fc fragment-binding activity of Mycoplasma salivarium cells was remarkably enhanced by trypsin treatment of the cells. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis profile of proteins of the cells treated with trypsin was the same as that of the cells treated with pronase, although pronase treatment had been shown to reduce the activity in our previous study (FEMS Microbiol. Lett. 123, 305-310, 1994). This contradiction was clarified by the finding that trypsin bound the Fc fragment more strongly than the cells, and a small amount of trypsin remained in the cells treated with trypsin and washed well. On the basis of these results, it was concluded that the enhancement of cell activity by trypsin treatment was ascribed to binding of the Fc fragment to trypsin remaining in the trypsin-treated cells.
