    Abstract

    So far, the sol-gel process has been available to prepare precursor gels of bioactive glasses with various compositions. In this report, we described a novel coating method of bioactive gels on a titanium substrate where the sol-gel transition is controlled by applying external electric fields. The application of a constant current of 10 mA/cm2 in an acidic sol containing pre-hydrolyzed tetraethoxysilane, calcium nitrate, and ammonium dihydrogen phosphate led to the deposition of gels on the titanium cathodes due to the generation of OH by water electrolysis as a catalyst of the sol-gel transition. The obtained gels, which were characterized to be amorphous and consisted of Si, Ca, and P, covered the titanium substrates as a coating. The bioactivity of the gels deposited was confirmed by soaking in a simulated body fluid (SBF) up to 7 days, suggesting that the electrochemically assisted sol-gel process is promising for providing bioactive coatings on metallic implants.

    To improve the cytocompatibility of mineral trioxide aggregate (MTA) cement and its ability for reparative dentin formation, the effect of adding choline dihydrogen phosphate (CDHP), which is reported to be biocompatible, to MTA cement was investigated. The L929 cell proliferation showed that the addition of CDHP improved cell viability. The addition of CDHP shortened the setting time of MTA cement, with a significant decrease in consistency above 0.4 g/mL. Diametral tensile strength of the set cement was improved by the addition of 0.4 g/mL CDHP. Solubility was judged to be within the range of clinical application. The spontaneous precipitation of low crystalline hydroxyapatite was examined by immersing the set cement in phosphate buffer saline, and it was found that the ability of the cement with 0.4 g/mL of CDHP was significantly improved compared with that of the cement without CDHP.

    Natural calcium phosphates derived from fish wastes are a promising material for biomedical application. However, their sintered ceramics are not fully characterized in terms of mechanical and biological properties. In this study, natural calcium phosphate was synthesized through a thermal calcination process from salmon fish bone wastes. The salmon-derived calcium phosphates (sCaP) were sintered at different temperatures to obtain natural calcium phosphate bioceramics and then were investigated in terms of their microstructure, mechanical properties and biocompatibility. In particular, this work is concerned with the effects of grain size on the relative density and microhardness of the sCaP bioceramics. Ca/P ratio of the sintered sCaP ranged from 1.73 to 1.52 when the sintering temperature was raised from 1000 to 1300 °C. The crystal phase of all the sCaP bioceramics obtained was biphasic and composed of hydroxyapatite (HA) and tricalcium phosphate (TCP). The density and microhardness of the sCaP bioceramics increased in the temperature interval 1000–1100 °C, while at temperatures higher than 1100 °C, these properties were not significantly altered. The highest compressive strength of 116 MPa was recorded for the samples sintered at 1100 °C. In vitro biocompatibility was also examined in the behavior of osteosarcoma (Saos-2) cells, indicating that the sCaP bioceramics had no cytotoxicity effect. Salmon-derived biphasic calcium phosphates (BCP) have the potential to contribute to the development of bone substituted materials.

    Mechanochemically activated graphite (Gp) and iron particle surfaces were prepared, and the Gp/iron composite interfaces were elucidated to utilize new functions. Activated iron particles with the oxygen atoms prepared by a milling under oxygen atmosphere were shown. Alternately, the Gp particles activated by the milling under argon atmosphere easily agglomerated because of the dangling bonds on the edge surfaces. In the composites, the interfacial bonds (e.g., FeOC) between the Gp and iron oxides on the iron were clearly induced while preserving the carbon atom six-ring structures. Furthermore, the molecular orbital calculations suggested that the activated oxygen atoms in the iron oxides effectively abstract hydrogen atoms from the CH groups of the Gp edge, resulting in the active bare carbon atoms which can form the interfacial bonds; therefore, the interfacial FeOC bonds for a higher adhesive strength were successfully prepared.

    Iontophoresis (IOP) is a noninvasive method of delivering medication transcutaneously through the skin. The electrodes used in this method should tightly fit to rough and irregular surfaces and be biologically safe, easy to handle and prepare, and cost-effective. To satisfy these requirements, calcium alginate gel can be a candidate electrode for IOP. Using calcium alginate gel electrodes, we examined whether lidocaine can be effectively transported across an excised rat skin by squarewavealternating current (AC) application. A squarewave AC with either a 70 % or 80 % duty cycle was continuously applied to 0.5 % calcium alginate gel electrodes containing 10 % lidocaine at 10 V and 1 kHz for 60 min. Lidocaine concentration was measured using a spectrophotometer and the temperature of the gel was determined. The lidocaine concentrations for AC-IOP at the 70 % and 80 % duty cycles were significantly higher than that without AC-IOP. Furthermore, the group with the 80 % duty cycle showed higher lidocaine concentrations than the group with the 70 % duty cycle. The temperatures of all the groups were lower than 28 ℃ throughout the procedure. In conclusion, the calcium alginate gel can be used as a possible matrix for IOP electrodes.

    <title>ABSTRACT</title>B-type carbonate apatite (B-CAp) powders were prepared by a wet method using Ca(OH)2 suspension and H3PO4 solution including NaHCO3 as a carbonate source, and porous B-CAp ceramics with two different amounts of carbonate contents were fabricated by sintering freeze-dried mixtures of the powders and gelatin composite. The porous B-CAp ceramics prepared had three-dimensionally interconnected pores. The sinterability of B-CAp ceramics was dependent on the chemical composition, especially sodium content and vacancy of OH site, and the carbonate contents did not directly influence the dissolution rate of porous B-CAp ceramics.

    <title>ABSTRACT</title>Nitinol was coated with biocompatible calcium phosphate materials by pulsed electrolytic deposition (ELD) to reduce toxic metal-ions elution. The pulse ELD for the stents was carried out with changing the current off-periods (<italic>t</italic>off) of the pulse wave. The pulse ELD suppressed the generation of H2 gas due to the electrolysis of water on a calcium phosphate layer and improved the adhesiveness of the coating layer on nitinol compared with a conventional DC-ELD. The coating layers were identified to be octacalcium phosphate (OCP) at lower <italic>t</italic>off, while they were transformed to dicalcium phosphate anhydraous (DCPA) with an increase of <italic>t</italic>off. The layers of OCP or DCPA on the nitinol surface were subjected to a NaOH treatment at 60°C for 3days to transform them into hydroxyapatite (HAp). From results of a metal-ions elution test, the deposited calcium phosphates suppressed nickel ions elution at one quarter compared with the bare nitinol stent. These results indicate that the pulse ELD of biocompatible calcium phosphate materials on the nitinol stent was one of the best techniques to create firmly attached coating on it and reduce toxic nickel ions elution.

    <title>ABSTRACT</title>Hydroxyapatite-chondroitin sulfate (HAp/ChS) composites were synthesized with calcium hydroxide suspension and phosphoric acid solution containing ChS through a precipitation method, and the microparticles were then fabricated by a spray dry method with the suspension of the composites. Bovine serum albumin (BSA) with negative charge or lysozyme (LYZ) with positive charge at pH7.0 was adsorbed onto the HAp/ChS microparticles. However, the HAp/ChS microparticles adsorbed LYZ more than the HAp microparticles compared with BSA due to the electrostatic interaction from negatively-charged sulfate or carboxyl group of ChS in the composites. The release property of BSA from the HAp/ChS microparticles was evaluated in Dulbecco’s phosphate buffered saline (pH7.2). The HAp/ChS microparticles released quickly 100% of the adsorbed BSA, while HAp microparticles released 45% of BSA. These results indicated that incorporation of ChS in the microparticles controls the adsorption and release properties of protein due to the electrostatic interaction. The HAp/ChS microparticles therefore are a candidate of a carrier for drugs like vaccines.

    The fabrication of iron–graphite composites was investigated using a milling process. Based on the milling time of the process, the average particle size of the host iron particles decreased to 10 µm. The size of the graphite particles decreased to a nanometer scale, resulting in the graphite strongly adhering to the iron surfaces. Raman spectra revealed that the graphite aromatic ring structure disappeared and the iron near-surface layer was changed to iron oxide phases. The dangling bond of the fragmented graphite combined with the oxidized iron surface by interfacial binding to efficiently form the interfacial composites.

    This study is concerned with multilayered protein adsorption on carbonate apatite (CAp) that is measured with a quartz crystal microbalance (QCM) technique. A carbonate apatite-deposited QCM sensor was prepared by electrophoretic deposition (EPD). On the CAp-deposited QCM sensor, fibrinogen adsorption followed by thrombin adsorption was investigated. The adsorption of fibrinogen on CAp led to a clear decrease in the resonance frequency of the sensor, which means that fibrinogen adsorbed on the CAp-deposited QCM sensor. The adsorbed amount of fibrinogen was larger on CAp than on a pristine sensor (Au). Although successive thrombin adsorption occurred on fibrinogen adlayers on both CAp and Au, the adsorbed amount and its conformation were different depending on the surface.

    The fabrication of copper–graphite composites by a milling process was investigated using a centrifugal ball mill. The copper particles were homogeneously milled in a graphite vessel, and the reaction time was varied. Scanning electron microscopy images clearly revealed that a fragment of graphite ground by the copper particle adheres to the copper particle surface, indicating the formation of a copper–graphite composite. The composite graphite amount per 1 g of copper particles increased to 0.46 mg at the milling time 5 min, and subsequently decreased to 0.25 mg at 60 min, indicating the suitable milling-time for the interfacial adhesion. When using only the copper particles, the naturally oxidized layer on the surface decreased with milling. On the other hand, when using only the graphite, the characteristic graphite structure is disrupted and the defect structure increased with milling. Thus, the new copper surface generated by milling strongly reacts with the defect structure of the graphite. It is suggested that the interfacial bonding between the copper and graphite was attributed to a Van der Waals attraction and/or binding force due to oxygen atoms located at the interface.

    Controlling the balance of endothelial cells (ECs) and smooth muscle cells (SMCs) in blood vessels is critically important to minimize the risk associated with vascular implants. Extracellular matrix (ECM) plays a key role in controlling the cellular balance, suggesting a promising source of cell-selective peptides. To obtain EC- or SMC-selective peptides, we start by highlighting sequence differences found among ECM molecules as enriched targets for cell-selective peptides. We explored the EC- or SMC-selective performance of tripeptides that are specifically enriched only in collagen type IV, but not in types I, II, III, and V. Collagen type IV was chosen since it is the major ECM in the basement membrane of blood vessels, which separates ECs and SMCs. Among 114 collagen type IV-derived tripeptides pre-screened from in silico analysis, 22 peptides (19%) were found to promote cell-selective adhesion, as determined by peptide array. One of the best performing EC-selective peptides (Cys-Ala-Gly (CAG)) was mixed into an electrospun fine-fiber, a vascular graft material, for practical application. Compared to unmodified fiber, the CAG containing fiber surface was found to enhance adhesion of ECs (+190%) while limiting SMCs (-20%). These results are not only consistent with the hypothesis of ECM as a source of cell selective peptides, but also suggest a new genre of EC- or SMC-selective peptides for tissue engineering applications. Collectively, these findings favorably support the screening approach used to discover new peptides for these purposes.

    <title>ABSTRACT</title>Bisphosphonate (Bp) was adsorbed on the surface of crystalline calcium phosphates (CP); hydroxyapatite (HAp), octacalcium phosphate (OCP) and Dicalcium phosphate dehydrate (DCPD). The amount of Bp adsorbed was the largest for DCPD per unit surface area, while the amount was the largest for HAp per unit weight. The composites of Bp and amorphous calcium phosphate (ACP) were synthesized by titrating calcium acetate solution into phosphate buffer solution containing Bp. The amount of Bp doped in the composites was 366 μg / mg and was approximately 7 times larger than those of Bp adsorbed on the crystalline Calcium phosphates. TG-DTA measurements of a Bp-calcium and the composite indicated exothermic peaks due to Bp combustion, of which temperature were shifted to higher temperature for the composite. Bp in the composites was gradually released into phosphate buffered saline, while Bp was rapidly released into acetate buffer solution accompanied with the dissolution of ACP. This result suggests that the composite of Bp and ACP has potential for a drug-carrier releasing Bp in response to the condition of osteoclastic bone resorption.

    <title>ABSTRACT</title>When a biomaterial is implanted into the body, blood proteins adsorb on its surface and subsequently cells adhere via the protein adlayer. Thus, the understanding of protein adsorption and conformational change on the biomaterial surfaces is of great importance to control the biocompatibility such as antithrombotic properties and cell adhesion behaviors. In this study, we synthesized hydroxyapatite (HAp) and carbonate apatite (CAp) by a wet method. Then we successfully fabricated the HAp and CAp sensors for QCM-R by an electrophoretic deposition method. Adsorption behavior of proteins on the bone substitute material can be monitored by using these apatite sensors. Bovine serum albumin and fibrinogen were employed for the model proteins, and monitored the adsorption behavior on the HAp, CAp and reference gold (Au) sensors by the QCM-R technique. As a result, we revealed that fibrinogen and bovine serum albumin adsorbs on the gold surface by hydrophobic interaction, and adsorbs on the HAp and CAp surfaces mainly by electrostatic force. Besides, we revealed that fibrinogen adsorbs on the Au surface more rigid than on the HAp and CAp surfaces while bovine serum albumin adsorbs on the HAp and CAp surface more rigidly than on the Au surface.

    Calcium alginate gel has some unique properties, such as the capability to keep the drugs, bioadhesiveness, safety, and low cost. The purpose of this study is to determine whether calcium alginate gel can be used as a matrix of electrodes for iontophoresis (IOP). We measured the concentration of lidocaine transported from calcium alginate gels with various concentrations of alginic acid using an in vitro experimental cell with square-wave alternating current (AC) application. Temperature and pH changes were also determined during AC-IOP. The results revealed that lidocaine was released from calcium alginate gels at concentrations nearly 1.71-fold larger at 5 V, 60 min after AC application than in the case of passive diffusion. Lidocaine transport depended on the alginic acid concentration in the gels. Although there were slight increases in temperature and pH, chemical and thermal burns were not severe enough to be a concern. In conclusion, the calcium alginate gel can be used as a possible matrix for IOP electrodes.

    <title>ABSTRACT</title>Collagen fibril membranes (CFMs) with a high mechanical property were fabricated with a lateral face evaporation method, in which type I atelocollagen extracted from tilapia scales was used. The density and thickness of the CFM obtained were 0.51 ± 0.04 mg/cm3 and 50 ± 5 μm. The collagen fibrils in the CFM had a similar periodic stripped pattern of 67 nm with native collagen fibrils. The CFM was crosslinked in gaseous glutaraldehyde for different duration in order to increase the mechanical property. The crosslinking degrees of the CFMs analyzed by free amino groups gradually increased to 70.3 % against the exposure duration until 6 hours, and reached a plateau. The denaturation temperatures of the CFMs with the crosslinking degrees at 20.4 % to 43% were linearly increased from 49°C to 75°C. The tensile strength of the CFMs was slightly improved until the crosslinking degree at 33.3 % and then the tensile strength rapidly increased to be 68 MPa. It was suggested that a percolation phenomenon took place in the CFMs by crosslinking of collagen fibrils with polymerized GA molecules.

    Europium (III) (Eu3+)-doped nanoporous silica spheres were synthesized, and the states of Eu3+ ions in the silica framework structure were investigated. The ordered nanopores were preserved with the doping at the Eu3+ molar concentration to Si up to 10 mol%, and the O-Si-O and Si-OH groups in the structures were clearly rearranged with the doping, indicating the interaction of Eu3+ with the O atoms. The significant morphological changes in the spheres were observed with the doping. The photoluminescence spectral shapes due to the transitions of D-5(0)-F-7(1) and D-5(0)-F-7(2) were indicative of the presence of the Eu3+ in an environment of a low symmetry. It was found that the Eu3+ was located inside the silica framework to electrostatically interact with the environmental O atoms, which would prevent the aggregation among Eu3+ ions to show the efficient luminescence. Therefore, the interactions between the Eu3+ ions and silica framework structures in the spheres were successfully clarified. (C) 2011 Elsevier Inc. All rights reserved.

    Europium (III) (Eu3+)-containing nanoporous silicas were successfully synthesized by two different loading methods (adsorption or doping), and the states of the Eu3+ in the structure were investigated. The Eu3+-adsorption indicated that the Eu3+ interacts with hydroxyl groups on the pore surface and H2O molecules to enhance the quenching, whereas the Eu3+-doping indicated that the Eu3+ was located inside the structure framework that produced the efficient luminescence.     <title>ABSTRACT</title>The dissolution kinetics of type B carbonate apatite (CAp) disks sintered at
    different temperatures were investigated under acidic conditions similar to
    an osteoclastic desorption process in bone remodeling. The type B CAp disks,
    which were uniaxially pressed at 98MPa and sintered at temperatures of 600
    to 700 ºC, showed single crystalline phase and a high relative density of
    60-62 % compared to the stoichiometric density of 3.16g/cm3 of
    hydroxyapatite (HAp). The dissolution rate of type B CAp disks sintered at
    650 ºC was 3.5 times faster than that of HAp disks at 650 ºC. These results
    indicate that the Type B CAp sintered disks show potential use as a
    biodegradable material for artificial bone.

    <title>ABSTRACT</title>Porous scaffolds of alkaline-soluble collagen including nanocomposite
    particles of chondroitin sulfate and low crystalline hydroxyapatite for
    cartilage regeneration were fabricated by freeze-drying and thermal
    dehydration treatments; porous collagen scaffolds were also synthesized as a
    reference. The scaffolds were cross-linked using glutaraldehyde (GA) vapor
    treatment in order to enhance biodegradable resistance. Microstructural
    observation with scanning electron microscope indicated that the scaffolds
    with and without GA cross-linkage had open pores between 130 to 200 μm in
    diameter and well-interconnected pores of 10 to 30 μm even after
    cross-linkage. <italic>In vitro</italic> biodegradable resistance to
    collagenase was significantly enhanced by GA cross-linking of the scaffolds.
    All these results suggest that the GA cross-linked scaffolds consisting of
    collagen, chondroitin sulfate, and low crystalline hydroxyapatite have
    suitable microporous structures and long-term biochemical stability for
    cartilage tissue engineering.

    <title>ABSTRACT</title>γ-Aminopropyltriethoxysilane (γ-APS) was grafted on stainless-steel and titanium substrates, and subsequently alginic acid layer was immobilized on them. Their surfaces were characterized with X-ray photoelectron spectroscopy (XPS) and contact angle measurement. Blood compatibility of thus obtained substrates was evaluated in terms of both the number of the adhered platelets and blood clotting factors for plasma contacted with the substrates such as active partial thromboplastin time (PTT), prothrombin time (PT), and amount of fibrinogen (Fib). The steel and titanium substrates with alginic acid layer did not affect blood clotting factors. <italic>In vitro</italic> platelet adhesion assay indicated that those substrates adhered less number of platelets than non-treated substrates. Hence the alginic acid immobilization leads to blood compatible surfaces.

    イオントフォレシスは、イオン化した薬物などを電流によって経皮・経粘膜で体組織に導入する方法である。これは薬物の経口投与と比較して、(1)肝臓での初回通過効果の影響を受けない、(2)胃腸によるdegradationを受けない、(3)注射の痛みがない、(4)薬物の長時間投与が可能、などの特徴がある。しかしこのような電極では薬物の担持量や放出量は制御されておらず、長時間にわたる経皮投与の点で課題を残している。そこで本研究では、薬物の担持・放出が制御可能な高分子ゲル層を電極金属と接合させた電極材料の作製を目的とした。薬物担持層として正荷電の麻酔薬と相互作用するアルギン酸ナトリウムを用い、電場によるアルギン酸ゲル層の堆積と金属の接合性について報告する。

    The protein adsorption on the alginic acid layers covalently immobilized on stainless-steel and titanium was examined by Fourier transform infrared spectroscopy using a reflection adsorption method (FT-IRRAS). The relationship between platelet adhesion and protein adsorption was also investigated. The contact angle measurement and FT-IRRAS spectra confirmed the successful immobilization of γ-APS and alginic acid onto stainless-steel and titanium substrates. The result shows that less protein adsorption leads to blood-compatible surface on the alginic acid-immobilization.

    With the intent to combine osteoconductivity with angiogenic property, the present study aimed to fabricate copper and silicon co-doped β-tricalcium phosphates (β-TCP).
    Copper-doped β-TCP(s) were successfully synthesized via both solid-state and liquid-phase reaction. In addition, the effect of copper on the dissolution behaviors of copper TCP (CuTCP) was also investigated. The incorporation of copper in the β-TCP resulted in a decrease in the dissolution rate of CuTCP, which was caused by a rate-determining dissolution process of a disordered phase around the CuTCP.
