    The purpose of this study was to evaluate the mechanism of action and the inhibiting effects of two types of desensitizers against dentin demineralization using pre-demineralized hypersensitivity tooth model in vitro. In this study, we confirmed that a hypersensitivity tooth model from our preliminary experiment could be prepared by immersing dentin discs in an acetic acid-based solution with pH 5.0 for three days. Dentin discs with three days of demineralization were prepared and applied by one of the desensitizers containing calcium fluoro-alumino-silicate glass (Nanoseal, NS) or fluoro-zinc-silicate glass (Caredyne Shield, CS), followed by an additional three days of demineralization. Dentin discs for three days of demineralization (de3) and six days of demineralization (de6) without the desensitizers were also prepared. The dentin discs after the experimental protocol were scanned using swept-source optical coherence tomography (SS-OCT) to image the cross-sectional (2D) view of the samples and evaluate the SS-OCT signal. The signal intensity profiles of SS-OCT from the region of interest of 300, 500, and 700 µm in depth were obtained to calculate the integrated signal intensity and signal attenuation coefficient. The morphological differences and remaining chemical elements of the dentin discs were also analyzed using scanning electron microscopy and energy-dispersive X-ray spectroscopy. SS-OCT images of CS and NS groups showed no obvious differences between the groups. However, SS-OCT signal profiles for both the CS and NS groups showed smaller attenuation coefficients and larger integrated signal intensities than those of the de6 group. Reactional deposits of the desensitizers even after the additional three days of demineralization were observed on the dentin surface in NS group, whereas remnants containing Zn were detected within the dentinal tubules in CS group. Consequently, both CS and NS groups showed inhibition effects against the additional three days of demineralization in this study. Our findings demonstrate that SS-OCT signal analysis can be used to monitor the dentin demineralization and inhibition effects of desensitizers against dentin demineralization in vitro.

    This study was designed to evaluate the effects of recombinant human BMP-2 (rhBMP-2) injected into the palatal periosteal sites in Wister rats on bone formation. The implant sites were separately divided into four groups according to the dosage of rhBMP-2 : 0 μg, 0.1 μg, 0.5 μg and 1.0 μg. The rats were sacrificed at 3 weeks after implantation, followed by histopathologic observation and histometric evaluation of new bone. Implants had disappeared at every group and new bone formation was observed at 0.1 μg, 0.5 μg and 1.0 μg. New bone was continuous with the original bone at 0.5 μg and 1.0 μg. Thickness of new bone increased as the dosage increased from 0 μg to 0.5 μg (P<0.05), and did not significantly change as the dosage increased from 0.5 μg to 1.0 μg. These results suggested that rhBMP-2 injected into the palatal periosteal sites could lead to induce new bone formation, although its effects varied according to the dosage of rhBMP-2.

    The purpose of this study was to evaluate the effect of direct pulp capping using recombinant human BMP-2 (rhBMP-2) on dentin regeneration. RhBMP-2 (1.0 mg/ml) was dissolved in alginate-based gel (AG). Cavities with the exposed pulp area were prepared of first molars in Wister rats. All cavities were divided into two groups: The exposed pulps were covered with AG contained rhBMP-2 (BMP group), and covered with AG alone (AG group). Then they were sealed with glass ionomer cement (Fuji IILC EM). The animals were sacrificed at 3 and 8 weeks after operation. In both groups, the covered materials were observed at 3 weeks after operation, while they had disappeared and the tertiary dentin formation was observed into the prepared cavity at 8 weeks after operation. Thickness of the tertiary dentin of BMP group was significantly higher than that of AG group (P<0.05). These results suggest that AG is effective scaffold and direct pulp capping using AG contains rhBMP-2 is capable of enhancing dentin regeneration.

    This study was designed to examine the effects of dose of recombinant human BMP-2 (rhBMP-2) on bone formation at palatal sites in 10-week-old (10w) and 70-week-old (70w) rats, when combined with a polylactate-polyglycolate copolymer/gelatine sponge (PGS). New bone formation was observed at six weeks after implantation. In the 10w rats, thickness of new bone (TNB) increased as the dosage increased from 0 microg to 4 microg, and decreased significantly as the dosage increased from 8 microg to 24 microg. In contrast, in the 70w rats, TNB increased as the dosage increased from 0 microg to 16 microg, and did not significantly change as the dosage increased from 16 microg to 24 microg. These results suggest that the most effective dosage of rhBMP-2 for induction of bone formation varies according to age.

    Recombinant human bone morphogenetic protein (BMP)-2 is known to induce significant regeneration of bone, but the influence of osteoporosis on bone formation is not understood. The aim of this study was to investigate the influence of osteoporosis on bone formation by histopathological and histometrical evaluations. Five-week-old KO OCIF/Jcl (homo) mice (osteoporosis group) and C57BL/6NJcl mice (normal group) were used. Atelocollagen gel with 0, 1, 5, 10 and 15 μg/ml BMP-2 were prepared and injected into the periosteum of the mice's cranial bone. Histopathological observation and histometric measurement of new bone and bone density were performed. The Mann Whitney U test was used for statistical significance. Both groups increased new bone in a concentration-dependent manner. At BMP concentrations of 0 and 1μg/ml, neither group showed new bone, and at BMP of 5 and 15μg/ml, the osteoporosis group showed significantly more bone than the normal group (p<0.05). Bone density was significantly lower in the osteoporosis group than in the normal group at BMP 5,10 and 15μg/ml separately (p<0.05). As a result, it was concluded that the amount of new bone induced by BMP-2 increases and the bone density decreases in osteoporosis.

    We have developed and applied a new electronic portfolio system to educate 65 dental residents for postgraduate clinical training in Okayama University Hospital of Medicine and Dentistry since the 2006 academic year. The purpose of this study was to examine the situation of postgraduate clinical training concerned with conservative dental treatment by using our electronic portfolio system in order to improve the postgraduate clinical training program at our hospital. At first, the number of patients whom the dental residents had treated was examined. Next, the records related to conservative dental treatment that the residents had put in the electronic portfolio system were exported and divided into three groups: restorative treatment, endodontic treatment and periodontal treatment, and the details were analyzed. The average number of patients for the period of their internship was 17.0±4.9 persons and the total number of records in the electronic portfolio system was 23,911. The records concerned with conservative dentistry accounted for 47.3% (11,316 records) of the total. The frequency of the restorative, endodontic, and periodontal treatments were 9.4% (2,239 records), 9.8% (2,335 records), and 28.2% (6,742 records), respectively. Detailed analysis demonstrated that residents experienced over 10 cases of "root canal filling" and "scaling and root planing", and over 20 cases of "resin filling", "root canal enlargement and/or application", "periodontal tissue examination", "tooth brushing instruction", "sonic scaling", and "supportive periodontal therapy". The results demonstrated that the dental residents experienced conservative dental treatment with a frequency of over 50% (almost 50% of all dental treatments) during their postgraduate clinical training program in Okayama University Hospital of Medicine and Dentistry, and that they experienced over 10 cases of basic treatment procedures of conservative dentistry.

    <p> Purpose: Recently, the application of composite resin restorations has expanded. Although chromatic studies of composite resins have progressed, color differences measured by colorimeters and visual color evaluation do not always match. The aim of this study was to establish a method of evaluating the visual color of composite resins by using the visual analog scale (VAS), which evaluates subjective data as a continuous variable, and to examine its validity.</p><p> Method: One structural color composite resin (experimental resin : ECM-001) and three commercial composite resins of shade A3 were used in this study. Class Ⅴ and class Ⅲ cavities were prepared in artificial maxillary central incisors of four shades (A1, A2, A3, and A3.5) and each composite resin was filled and its color matching was evaluated on a black-and-white background using VAS. The correlation between VAS value and visual evaluation was analyzed. The color matching of teeth and composite resin were statistically analyzed using one-way analysis of variance and Tukey's HSD test (p<0.05). Background differences in the same sample were analyzed by the t-test (p<0.05).</p><p> Results: In the Class Ⅴ restoration, ECM-001 showed better color matching than other composite resins. In the Class Ⅲ restoration, ECM-001 showed excellent color matching with a wide range of teeth shades on the white background. The VAS values of the other composite resins were higher on the black background than on the white background. However, the VAS value of ECM-001 on the black background tended to decrease. Because the evaluation of VAS color matching by evaluators showed the same tendency, the VAS value reflects information that can be recognized by visual observation. It is suggested that the evaluation of VAS value correlates with visual evaluation.</p><p> Conclusion: It is suggested that the evaluation of color matching of composite resin restorations using VAS is useful for structural color composite resins, providing a new method of visually evaluating composite resin restorations.</p>

    <p> Purpose: Clinically, resin composite repair for dentin hypersensitivity with substantial defects such as wedge-shaped defects is commonly performed a few days after applying the densensitizer to dentin. In this study, we investigated the influence of pre-treatment of dentin densensitizers on bond strength.</p><p> Materials and Methods: We applied three desensitizers (MS coat ONE, F Varnish, and Nanoseal) on an artificial model of hypersensitive dentin using human premolars, and observed the surface structure by scanning electron microscope (SEM). Then, the specimens to which the three desensitizers had been applied were treated with Clearfil Mega Bond FA. We morphologically observed by SEM the surface of dentin which had been treated with the primer. Furthermore, the micro-tensile bond strength (μTBS) for the specimens which were incrementally built up by resin composites was measured in a universal testing machine. The interface between resin and dentin of specimens was observed by SEM and confocal laser scanning microscope (CLSM).</p><p> Results: The surfaces of specimens to which each desensitizer had been applied showed morphological characters having the effects of suppressing hypersensitivity. Regarding μTBS, there were no significant differences among all groups. In the group of MS coat ONE, MS coat ONE was not completely removed by the application of Mega Bond FA primer. Although the penetrations of the primer into dentin tubules were small, the thickness of the bond layer was greater. The specimens of F Varnish showed the functions of self-etching adhesive systems, because the varnishes had been partially removed by Mega Bond FA primer. In the group of Nanoseal, Mega Bond FA primer was almost removed by Nanoseal, and penetrated deeply into the dentinal tubules.</p><p> Conclusion: The three desensitizers used in this study had no effect on the bond strength of self-etching adhesive systems to the artificial model of hypersensitive dentin.</p>

    &emsp;Purpose: By incorporating sodium fluoride in adhesive resin cement, the release of fluoride from the cement is expected to strengthen the teeth and prevent secondary caries. However, there is a concern that such fluoride incorporation and release could impair the physical properties of the cement. In this study, we investigated the influence of incorporating sodium fluoride on the flexural strength, water absorption and solubility of adhesive resin cement.<br>&emsp;Materials and Methods: The experimental resin cements were prepared at concentrations of 0-20 wt% sodium fluoride. The amounts of fluoride released from the cements were measured from day 1 to day 84. Next, a three-point bending test of the experimental resin cements was performed after initial curing (24 hours storage in water at 37&deg;C) and after thermal cycle loading (5&deg;C-55&deg;C, 10,000 times). Furthermore, water absorption and solubility following 30 days immersion in water were measured.<br>&emsp;Results: Fluoride release increased with an increase in the content rate of sodium fluoride. After initial curing, the three-point bending strengths of 5 wt% and 10 wt% sodium fluoride compounded cement were not statistically decreased compared to those of sodium fluoride 0 wt%, but were significantly reduced by incorporation of more than 15 wt%. There were no significant differences in the strength between 15 wt% sodium fluoride compounded cement and 20 wt% sodium fluoride compounded cement. Similar results were also observed in the comparison of flexural strength after thermal cycle loading. Regarding the 30-day water absorption amount, there was no significant difference between 5 wt% sodium fluoride compounded cement and that of 0 wt% sodium fluoride cement. However, the 30-day water absorption of 10 wt% sodium fluoride compounded cement was significantly increased compared to that of 0 wt% sodium fluoride compounded cement. The solubility was not significantly different between the content rate of 0-10 wt% sodium fluoride.<br>&emsp;Conclusion: Increasing the sodium fluoride content improved the amount of fluoride release. On the other hand, the flexural strength of the cement was decreased, and the amounts of water absorption and solubility were increased. Within the limit of this study, it is suggested that 10 wt% sodium fluoride compounded cement could provide fluoride release without decreasing mechanical strength.

    Purpose: The purpose of this study was to evaluate the effects of low-intensity pulsed ultrasound (LIPUS) on bone formation induced by polylactate-polyglycolate-copolymer/gelatin sponge complexes (PGS) containing recombinant human bone morphogenetic protein-2 (rhBMP-2) at palatal subperiosteal sites (n=24) in 15-week-old male Wistar rats. Methods: RhBMP-2/PGS (1.0 μg/μl) was implanted into the palatal subperiosteal site and treated with LIPUS (1.0 MHz, 240 mW, 5 min) every 3 days (LI-BMP group) or without LIPUS (BMP group). In addition, rhBMP-2/PGS (1.0 μg/μl) was not implanted and rats were treated with LIPUS (LI group) or were not treated with LIPUS (Cont group). Animals were euthanized 3 weeks after surgery. Results: Thickness of new bone in LI-BMP and BMP groups was significantly higher when compared with LI and Cont groups (p<0.05). There were no significant differences in thickness of new bone between LI-BMP and BMP groups (p>0.05). Newly formed bone in the BMP group was continuous with the original bone. On the other hand, newly formed bone was largely integrated with the original bone in the LI-BMP group. Conclusion: These results suggest that LIPUS has the ability to enhance maturation of bone induced by rhBMP-2 in the case of palatal subperiosteal implantation in adult rats.

    Tissue regeneration is the important issue in the field of operative dentistry. We demonstrated in series of studies that human bone marrow mesenchymal stem cells (hMSCs) can differentiate into odontblasts under the certain conditions (Izawa S et al., 2009). Mesenchymal stem cells can differentiate into multiple types of cells derived from mesenchyme (Liechty KW et al., 2000). At the same time odontblasts are primarily derived from mesenchyme; thus, we expected that mesenchymal stem cells can differentiate into odontblasts.The results in series of our study indicated that TNFα is necessary for hMSCs to differentiate into odontblasts and suggest that TNFα activates potential totipotency of hMSCs and the activation can lead to the regeneration of target cells or tissues.

    The purpose of this study was to evaluate the effect of the method of removing carious dentin on resin-composite bond strengths to dentin using three self-etching adhesive systems: G-Bond (GC), One-up Bond F (Tokuyama Dental) or Clearfil Mega Bond (Kuraray Medical). The bur-cut, Er: YAG-lased and ultrasonic vibrated dentin was subjected to micro-tensile bond testing and observation of ultra-structure by scanning electron microscopy. A smear layer was not formed on the dentin surface after removing carious dentin in the case of Er: YAG-lased and ultrasonic vibrated dentin. Clearfil Mega Bond showed significantly higher micro-tensile bond strength to bur-cut and ultrasonic vibrated dentin compared with Er: YAG-lased dentin (p<0.05). One-up Bond F showed significantly higher micro-tensile bond strength to ultrasonic vibrated dentin compared with bur-cut and Er: YAG-lased dentin (p<0.05). It is suggested that the method of removing carious dentin by an ultrasonic vibration device is useful for dentin adhesion when using self-etching adhesive systems.

    The mineralization accelerating adhesion materials that added EVA+C (RMSB) were produced experimentally, and we studied it about a bond strength and the biocompatibility. As a result, bond strength decreased with increase of the EVA+C addition ratio. But the level of the decrease was inhibited when SBP30 was treated. The RMSB group showed a tendency to growth similar to Control at the early phase, and cell count was higher than an SB (Superbond) at high ratio of EVA+C group. It was a combination of SBP30 and 7P (SB/EVA+C = 40wt%/60wt%) that showed a high bond strength and a good cell proliferation. Also, as a result of animal experiment, 7P had weaker inflammation than an SB and mineralized tissue formation was good. It was suggested that it was obtained high adhesive property and biocompatibility by adding EVA+C in an SB at the appropriate ratio in conclusion.

    Dentin regeneration to the lost dentin is the supreme therapy to keep functionality, aesthetics and so forth in operative dentistry. In 2006, we demonstrated the human bone marrow mesenchymal stem cells (hMSCs) differentiated into odontoblast by the stimulation of Emdogain Gel and TNF-&alpha; set. In 2008, we tried to demonstrate odontoblast that differentiated from hMSCs generate dentin under the artificially synthesized appropriate scaffold and molecular signal in vitro. As a result, the differentiated cell tissue tested positive for osteocalcin by fluorescence antibody method and enzyme antibody technique and positive for calcium compounds with the alizarin red S staining. In addition, we detected the dentin-like tissue outside the cell walls by phase contrast microscopy. These results suggested that hMSCs differentiate into odontoblast and generate dentin using Emdogain Gel and TNF-&alpha; set and peptide hydrogel.

    The purpose of this study was to evaluate the effect of delayed application of the dropped all-in-one adhesives (Clearfil tri-S Bond and G-Bond) on enamel and dentin bond strengths. Human extracted premolars were used and flat enamel and dentin surfaces ground by 600-grit SiC paper were prepared. The tooth surfaces were treated with Clearfil tri-S Bond and G-Bond which were left for 0,5,10 and 30min after dropping of a adhesives on a shaded dish at constant temperature and humidity, and then covered with resin composites according to the manufacturers' instructions. After immersion in 37℃ water for 24 h, the teeth were serially sectioned into multiple slices and the micro-tensile bond strength was measured in a universal testing machine at a crosshead speed of 1.0mm/min. Data were statistically analyzed using one-way ANOVA and Dunnett's test at a significance level of 0.05. The weight changes of the dropped adhesives were also measured until 30 min under the shaded condition. In the comparison of the leaving time, a significant difference in the bond strength was only detected between 0 min and 30 min on enamel. The bond strength of Clearfil tri-S Bond to enamel after 30 min increased, but that of G-Bond after 30 min decreased (p<0.05). There was no significant difference in the bond strengths of Clearfil tri-S Bond and G-Bond to dentin. The weights of Clearfil tri-S Bond and G-Bond decreased with the leaving time after dropping and the percentage of the weights of adhesives were 82.7% for Clearfil tri-S Bond and 52.0% for G-Bond after 30min. These results suggested that the long-term leaving after dropping of the all-in-one adhesives affected the bond strengths to enamel bv evanoration of the solvents in adhesives.

    Dopamine (DA) is a major central nervous system (CNS) neurotransmitter with many important physiological activities. Investigations into the neuroanatomy and neurologic functions of the dopaminergic neural systems have generated much debate. Regarding neuroanatomy, physiological and pharmacological criteria have divided DA receptors into D1 and D2 subtypes. The genes encoding these subtypes have been cloned and classified into a D1 subfamily encompassing D1 and D5 receptors and a D2 subfamily with D2, D3, and D4. Based on the sequences of the cloned receptors, we prepared antibodies and riboprobes to elucidate the expression of the corresponding proteins and mRNAs in the rat area postrema (AP) by immunohistochemistry and in situ hybridization (ISH). The AP was obtained from adult male Sprague-Dawley rats undergoing brain surgery, and tissue samples were used for RT-PCR, immunohistochemistry, and ISH. The results showed that D2 and D5 receptors and their mRNAs exist in the rat AP. On the other hand, D1, D3, and D4 receptors and their mRNAs were not detected.

    This study investigated the bond strengths of one-or two-step self-etching adhesive systems to human sound, demineralized and remineralized dentin. Remineralized dentin was created by remineralization for1or4weeks after demineralization. The dentin surfaces were treated with Clearfil SE Bond or Clearfil Tri-S Bond and the microtensile bond strengths were measured. There were no significant differences in the bond strength of Clearfil SE Bond among the three groups (demineralized, 1-week and4-week remineralized dentin groups) except for the sound dentin group. On the other hand, the bond strength of Clearfil Tri-S Bond to1-week remineralized dentin was significantly higher than those to demineralized dentin and4-week remineralized dentin. In comparing the two adhesive systems, Clearfil SE Bond produced higher bond strengths to all dentin surfaces than Clearfil Tri-S Bond. In conclusion, it is suggested that the difference of the adhesive system affected the bond strength to demineralized and remineralized dentin.

    This study investigated the effect of light-curing of Clearfil DC Bond (DC Bond) on the micro-tensile bond strength of Clearfil Esthetic Cement to dentin. Extracted human teeth were used in this study. The coronal dentin surfaces were prepared, and treated with DC Bond with or without light-curing, or ED primer II according to the manufacturer's instruction and luted composite block using Clearfil Esthetic Cement. The bond strengths were measured with the micro-tensile bond test. The bond strengths were statistically compared with ANOVA and Duncan's new multiple range tests. The debonded specimens and resin-dentin interfaces were also examined by SEM. There were no significant differences in bond strength among all the groups. In the SEM observations, no morphological differences were found among all the groups. For all fractured surfaces in the debonded DC Bond with light-curing group, cohesive failure in resin was observed. Adhesive fracture was predominantly observed in the debonded DC Bond without light-curing group. The cohesive failure in resin was predominantly observed in the debonded ED primer group.<BR>In conclusion, although the type of fractured surface for the DC Bond without light-curing group differed from that for the DC Bond with light-curing group, there was no significant difference in the dentin bond strength of Clearfil Esthetic Cement between the two groups.

    This study was designed to examine the effects of bone formation induced by recombinant human BMP-2 (rhBMP-2), which was implanted together with a poly-lactate-polyglycolate-copolymer/gelatine sponge (PGS) at palatal sites in 30-week- old male Wister rats. The rats were divided into four groups according to the dose of rhBMP-2: 0&micro;g, 4&micro;g, 8&micro;g and 16&micro;g.<BR>New bone formation was observed in every group, and new bone was continuous with the original bone at 6 weeks after implantation. Thickness of new bone (TNB) increased as the dose increased from 0&micro;g to 8&micro;, reached the maximal level at 8&micro;g. In contrast, TNB decreased significantly as the dose increased from 8&micro;g to 16&micro;g.<BR>The results of this study indicate that new bone formation varies according to the dose of rhBMP-2 at palatal sites in adult rats.

    The aim of this study was to investigate the effect of bioabsorbable material on the direct pulp capping to know whether this material has a function in dentin regeneration by bioabsorption.<BR>Class V cavities with the exposed pulp area were prepared on the buccal surface of first molars in young beagle dogs. All cavities were conditioned with a self-etching primer (Clearfil SE Primer). In the cavities of right side (experimental group), the exposed pulp was covered with a bioabsorbable sheet (Kurabio AG), and then the cavities were coated with an adhesive agent (Clearfil SE Bond) and resorted with a resin composite. The left side cavities (control group) were applied the adhesive agent without the bioabsorbable sheet and restored with a resin composite. On three and six months after surgery, the teeth were extracted, immersion-fixed and decalcified. Afterward, the teeth were frozen-sectioned, stained with hematoxylin and eosin and observed using a light microscopy.<BR>In the experimental group, a large amount of the tertiary dentin was formed into the prepared cavity. On the other hand, the tertiary dentin was almost not formed into the cavity in the control group.<BR>The significantly tertiary dentin formation was recognized into the prepared cavity when the exposed pulp area had been covered by a bioabsorbable material. This result suggested that the space making by a bioabsorbable material could produce the derivation of pulp tissue and then dentin regeneration into the pre-pared cavity.

    The purpose of this study was to evaluate the effect of bone formation induced by recombinant human BMP-2 (rhBMP-2) combined with bioabsorbable material Kurabio&reg;AG (AG) at palatal subperiosteal sites in Wistar rats. AG was freeze-dried alginated gel dressing.<BR>Experimental sites were divided into three groups according to the implans : BMP group (rhBMP-2 combined with AG), AG group (AG alone) and Control group.<BR>In every group, infected region or atypical cell proliferation was not observed, and new bone formation was observed at 6 weeks after implantation. Thickness of new bone (TNB) of BMP group was significantly higher than that of AG and Control groups. There was no significant difference in TNB between AG and Control groups.<BR>These results suggest that rhBMP-2 combined with AG have the ability to promote bone formation, although AG alone do not have any effect on bone formation.

    We studied bone formation induced by recombinant human bone morphogenetic protein-2 (rhBMP-2) combined with polylactate-polyglycolate-copolymer/gelatine sponge complexes (PGS) at palatal subperiosteal sites in 54 male Wister rats with STZ-induced diabetes mellitus (DM), insulin-treated STZ-induced diabetes mellitus (INS-DM), or nondiabetes mellitus (nonDM). Rats were divided into 6 groups consisting of PGS implanted alone (nonDM-PGS, DM-PGS, and INS-DM-PGS) and rhBMP-2 combined with PGS implanted (nonDM-BMP, DM-BMP, and INS-DM-BMP). Rats were sacrificed 6 weeks after implantation and histopathologically observaed and histometrically evaluated. New bone formation was observed in all groups, and the new bone was almost completely continuous with the original palatal bone. The DM-PGS group showed significantly less new bone thickness among the 3 groups with PGS implantion alone. NonDM-BMP, DM-BMP and INS-DM-BMP showed significantly greater new bone thickness than did nonDM-PGS, DM-PGS, and INS-DM-PGS groups.<br>These results indicate that rhBMP-2 combined with PGS enhances new bone formation continuous with original bone under STZ-induced diabetes status, although it decreases reparative bone formation.

    This study was designed to examine the effect of aging on bone formation induced by recombinant human bone morphogenetic protein-2 (rhBMP-2) combined with a fibrous collagen membrane (FCM). Implantation was done subperiosteally in bilateral palatal grooves in 34 male Wistar rats divided into three age groups: a 10-week-old group(10w group), a 30-week-old group(30w group) and a 70-week-old group (70w group). RhBMP-2-combined FCMs were implanted on the left palatal grooves as BMP-implanted sites (BMP site), while rhBMP-2 was not implanted on the right palatal grooves as control sites. The rats were sacrificed 6 weeks after implantation, and histometric evaluations were performed, New bone formation was observed in every site of each age group and the new bone was almost completely continuous with the original bone. The new bone volume (NBV) of the BMP site was significantly higher than that of the control site in each age group, The NBV of both the control and BMP sites were highest in the 10w group and lowest in the 70w group. The disparity of NBV between the control and BMP sites, which indicated the response to implanted BMP excluding the effect of skeletal growth and surgical stimulation. did not significantly differ among the age groups. These results indicate that rhBMP-2-combined FCM has the ability to induct new bone Formation continuous with original bone even in senescent rats. Furthermore, it appeared that, in the case of palatal subperiosteal implantation, the responsiveness to implanted BMP was independent of age, although the total volume of newly formed bone declined with aging.

    This study was designed to examine the aging effect on bone formation induced by recombinant human BMP-2 (rhBMP-2) combined with polylactate-polyglycolate-copolymer/gelatine sponge complexes (PGS) at palatal subperiosteal sites (N=72) in 36 Wister strain male rats, at 10 weeks, 30 weeks and 70 weeks of age. The rats at each age were divided into three groups: no implant (CN), PGS implant alone (CI), rhBMP-2 combined with PGS implant (E). The rats were killed at 6 weeks after implantation, and histopathologic observation and histometric evaluation were performed. In histopathologic observation, new bone formation was observed in every group at each age, and the new bone was almost completely continuous with the original palatal bone. In histometric evaluation, the new bone volume was largest in 10-week-old rats and gradually decreased by age in each group. At each age, the new bone volume in group E was significantly larger than that of groups CN and CI. These results indicate that rhBMP-2 combined with PGS induces new bone formation that is continuous with original bone not only in young rats but also in old rats which have less bone formation activity, although the volume of the new bone at the BMP-implant site shows gradual decrease by aging.     This study was designed to evaluate the effects of osteoporosis on hard tissue formation using growth factor(BMP-2)and low-intensity pulsed ultrasound (LIPUS)in aged or ovariectomy (OVX) rats. The experimental sites were divided into two groups: implantation of biodegradable gelatin hydrogel (Gel) containing BMP-2(BMP group) and Gel only (Gel group).In the aged rats, the new bone formation in the BMP group was significantly increased as compared with the Gel group, and the formation was promoted by using LIPUS. In the OVX rats, the BMP group was significantly increased compared to the Gel group, but the bone mineral density was decreased compared to the non-OVX rats. On the other hand new bone formation was promoted and almost integrated with the original bone by using LIPUS.
    These results suggest that growth factor and ultrasound therapy have some effects on hard tissue formation in the case of osteoporosis.

    This study evaluated the effects of aging on hard tissue formation induced by BMP using Low-intensity pulsed ultrasound(LIPUS)in young and aged rats.
    Tertialy dentin formation was promoted by using pulp capping material contained BMP when LIPUS treatment was performed. There was no significant difference between the young and the aged rat in the volume of tertialy dentin. Newly formed bone was promoted by implantation of material contained BMP at palatal subperiosteal site when LIPUS treatment was performed, although the volume of bone decreased with aging. These results suggest that the effects of aging on hard tissue formation might vary with the experimental sites.

    The purpuse of this study was to evaluate the effects of dentin regeneration(formation) using growth factor (BMP; Bone Morphogenetic Protein) and low-intesity pulsed ultasoud(LIPUS) on direct pulp capping. Tertiary dentin formation was promoted by using pulp capping material contained BMP in comparison with non-BMP. Furthermore, it was enhanced by the application of LIPUS.
    These results suggest that the combination of growth factor (BMP) and LIPUS has the ability to enhance dentin formation in the case of direct pulp capping.

    う蝕、歯周病に次ぐ第3の歯科疾患として注目されているTooth Wearは実に成人の80%以上が罹患していると言われており、大きく咬耗・摩耗・酸蝕症の3つに分類されている。さらに近年、Tooth surface loss(TSL,歯質表面損失症候群)という非う蝕性の表面歯質アパタイトの慢性的な損失を示した概念が発表された。TSLにはTooth Wearのほかにアブフラクションや歯根露出も含まれ、国民の注目を集めている。H21年度には、デジタルマイクロスコープを用いてヒト抜去歯の咬耗面を観察し、その表面積を測定した。H22年度には、人工酸蝕歯を用いて唾液による再石灰化能を検討するとともに、ワンステップボンディングシステムの人工酸蝕象牙質への接着性を検討した。健全ヒト抜去大臼歯の歯冠部咬合面象牙質平坦面を露出後、コーラ系炭酸飲料に5分間浸漬し、その後、ヒト唾液中に浸漬し、3分後および5分毎にヌープ硬さを計測しながら45分後まで測定した。コーラ浸漬後の露出象牙質面の硬さを100とした場合、唾液浸漬3分後には、ヌープ硬さが有意に上昇し、45分後には硬さは112まで上昇し、ヌープ硬さの平均値は健全部と同等の62.6を示した。また微小引張試験の結果から健全象牙質に対し、ビューティーボンドは35.5±1.51MPa(n=10)を示したのに対して、人工酸蝕象牙質では27.1±1.86MPaと、有意に低い値を示した。ヒト抜去歯の象牙質露出面は、コーラなどの酸性飲料により大きくヌープ硬さが減弱することが示されたが、唾液浸漬45分でほぼ健全な硬さにまで回復することが示された。この結果は、歯根露出した患者の歯磨き方法に大きな示唆を与えると考えられる。また、現在、疫学倫理委員会の承認の下に、デジタルマイクロスコープを用いた咬耗歯の疫学調査を継続中である。

    The purpose of this study was to evaluate the effect of dentin and bone regeneration using growth factor and ultrasound. As a result, the dentin formation was promoted by using the growth factor in the case of direct pulp capping. Newly formed bone was observed at subperiosteal sites using growth factor. Furthermore, the maturation of newly formed bone was enhanced by using ultrasound. These results suggest that growth factor and ultrasound have the ability to enhance hard tissue formation.

    The mineralization accelerating adhesion materials that added EVA+C (RMSB) were produced experimentally, and we studied the bond strength and biocompatibility. Bond strength decreased with increase of the EVA+C addition ratio. The RMSB group showed a tendency to growth similar to Control at the early phase, and cell count was higher than an SB (Superbond) at high ratio of EVA+C group. SBP30 and 7P (SB/EVA+C=40wt%/60wt%) showed a high bond strength and a good cell proliferation. Invivo, 7P had weaker inflammation than an SB and mineralized tissue formation was good. High adhesive property and biocompatibility was oftained by RMSB-7P.
