    The postharvest properties of two ultra-late maturing peach cultivars, “Tobihaku” (TH) and “Daijumitsuto” (DJ), were investigated. Fruit were harvested at commercial maturity and held at 25°C. TH exhibited the characteristics of normal melting flesh (MF) peach, including rapid fruit softening associated with appropriate level of endogenous ethylene production In contrast, DJ did not soften at all during 3 weeks experimental period even though considerable ethylene production was observed. Fruit of TH and DJ were treated with 5,000 ppm of propylene, an ethylene analog, continuously for 7 days. TH softened rapidly whereas DJ maintained high flesh firmness in spite of an increase in endogenous ethylene production, suggesting that DJ but not TH lacked the ability to be softened in response to endogenous and exogenous ethylene/propylene. DNA-seq analysis showed that tandem endo-polygalacturonase (<italic>endoPG</italic>) genes located at <italic>melting flesh (M)</italic> locus, <italic>Pp-endoPGM</italic> (<italic>PGM</italic>), and <italic>Pp-endoPGF</italic> (<italic>PGF</italic>), were deleted in DJ. The <italic>endoPG</italic> genes at <italic>M</italic> locus are known to control flesh texture of peach fruit, and it was suggested that the non-softening property of DJ is due to the lack of <italic>endoPG</italic> genes. On the other hand, TH possessed an unidentified <italic>M</italic> haplotype that is involved in determination of MF phenotype. Structural identification of the unknown <italic>M</italic> haplotype, designated as <italic>M</italic><sup>0</sup>, through comparison with previously reported <italic>M</italic> haplotypes revealed distinct differences between <italic>PGM</italic> on <italic>M</italic><sup>0</sup> haplotype (<italic>PGM-M<sup>0</sup></italic>) and <italic>PGM</italic> on other haplotypes (<italic>PGM-M<sup>1</sup></italic>). Peach <italic>M</italic> haplotypes were classified into four main haplotypes: <italic>M</italic><sup>0</sup> with <italic>PGM-M<sup>0</sup></italic>; <italic>M</italic><sup>1</sup> with both <italic>PGM-M<sup>1</sup></italic> and <italic>PGF</italic>; <italic>M</italic><sup>2</sup> with <italic>PGM-M<sup>1</sup></italic>; and <italic>M</italic><sup>3</sup> lacking both <italic>PGM</italic> and <italic>PGF</italic>. Re-evaluation of <italic>M</italic> locus in association with MF/non-melting flesh (NMF) phenotypes in more than 400 accessions by using whole genome shotgun sequencing data on database and/or by PCR genotyping demonstrated that <italic>M</italic><sup>0</sup> haplotype was the common haplotype in MF accessions, and <italic>M</italic><sup>0</sup> and <italic>M</italic><sup>1</sup> haplotypes were dominant over <italic>M</italic><sup>2</sup> and <italic>M</italic><sup>3</sup> haplotypes and co-dominantly determined the MF trait. It was also assumed on the basis of structural comparison of <italic>M</italic> haplotypes among <italic>Prunus</italic> species that the ancestral haplotype of <italic>M</italic><sup>0</sup> diverged from those of the other haplotypes before the speciation of <italic>Prunus persica</italic>.

    Split-pit in peach fruit is a problematic disorder. Split-pit fruit cannot be detected based on external appearance, and contamination of fruit by split-pit reduces its reliability in the marketplace. Here, we demonstrate that split-pit fruit can be identified by a nondestructive acoustic vibration method and a unique approach based on the ratio of the third (f(3)) to the second (f(2)) resonant frequency. The response-resonant frequency spectra showed that the peaks of f(2 )frequencies in split-pit fruit were shifted to much lower values than those in normal fruit, whereas those of f(3) frequencies showed only small shifts. The calculated f(3)/f(2 )ratios in most normal fruit were in the range of 1.35-1.4, whereas those in split-pit fruit were 1.45-2.0. Analysis of more than 300 fruit samples revealed that by setting the f(3)/f(2) cut-off value at &gt;1.45, 95% of split-pit fruit in the fruit samples were detected, whereas only 1.5% of normal fruit were missorted as split-pit fruit. A model for simulating the vibration properties of peach fruit was developed by using the finite element method.     Reddish pulp is one of the most serious disorders in peach production, in which originally white flesh undergoes pink to red discoloration with the development of a mealy texture. In order to clarify the relationship between the incidence of reddish pulp and flowering date, the occurrence of reddish pulp was investigated in 'Benishimizu' and 'Shimizu Hakuto' fruit with different flowering dates for three seasons. In both cultivars, the percentage of fruit with reddish pulp was higher in early flowering fruit than late flowering fruit in all three seasons. Analysis of pectin and boron contents in immature fruit sampled from June 23 to July 8, prior to observing the symptoms of reddish pulp, showed no statistically significant differences in pectin content between early and late flowering fruit. On the other hand, the boron content in flesh was significantly lower in early flowering fruit that bloomed on April 1 than in late flowering fruit that bloomed on April 11 in 'Benishimizu'. In addition, a significantly higher total polyphenol content, which is one of the characteristics of reddish pulp, was found in early flowering fruit. The results indicate that the flowering date has a large impact on the incidence of reddish pulp, and the disorder is more prevalent in early than late flowering fruit. The possible relationships among the development of reddish pulp, boron deficiency, pectin content, and increase in total polyphenol content are discussed.

    <p>We evaluated the water status of peach, grape, Japanese pear, and satsuma mandarin trees using a 'water-stress indicator sheet'. This sheet indicates the water status of a sample by changing color (from blue to light pink); this is caused by a reaction between the sheet and water evaporating from the leaf onto which it is pressed. Decreases in the evaporation rates of grapes, peaches, and Japanese pear under water-stress conditions can be estimated using the time required for the indicator sheet to change color. This color change occurs in 100–110 s when in contact with the leaves of non-water-stressed plants, but when the plant is under sufficient water stress to reduce the evaporation rate by half, the change in color takes 200 s. In satsuma mandarin, it takes 130 and 230 s, respectively. Thus, the time elapsing between initial contact and the change in sheet color can function as a measure of the water status. However, when taking measurements for practical use, it is necessary to maintain conditions such as solar irradiance at a constant level.</p>

    Metabolomic analysis revealed that Methylobacterium cells accumulate a large amount of ergothioneine (EGT), which is a sulfur containing, non-proteinogenic, antioxidative amino acid derived from histidine. EGT biosynthesis and its role in methylotrophy and physiology for plant surface-symbiotic Methylobacterium species were investigated in this study. Almost all Methylobacterium type strains can synthesize FGT. We selected one of the most productive strains (M. aquaticum strain 22A isolated from a moss), and investigated the feasibility of fermentative EGT production through optimization of the culture condition. Methanol as a carbon source served as the best substrate for production. The productivity reached up to 1000 mu g/100 ml culture (1200 mu g/g wet weight cells, 6.3 mg/g dry weight) in 38 days. Next, we identified the genes (egtBD) responsible for EGT synthesis, and generated a deletion mutant defective in EGT production. Compared to the wild type, the mutant showed better growth on methanol and on the plant surface as well as severe susceptibility to heat treatment and irradiation of ultraviolet (UV) and sunlight. These results suggested that EGT is not involved in methylotrophy, but is involved in their phyllospheric lifestyle fitness of the genus in natural conditions.

    ブドウ'ピオーネ'について、休眠の導入と覚醒の過程における芽の生理的変化を調査した。休眠導入期の8月から覚醒初期の12月まで露地で栽培されている個体から芽を採取し、呼吸量、エチレン生成量および含水率を測定した。調査期間を通して呼吸量は低く、エチレンもほとんど検出されなかった。芽の含水率は8月から10月まで僅かに上昇し、その後は変化がみられなかった。休眠覚醒初期の12月から発芽期の4月中旬まで、芽の呼吸量とエチレン生成量を測定した。呼吸量は4月上旬までは低く推移し、発芽(4月13日)の直前に急上昇した。エチレンは測定期間を通して低かった。休眠期の12月、1月および2月に採取した穂木を2%H2CN2または蒸留水(対照)で処理し、25℃以上に保ったプラスチックハウスに入れて発芽を調査するとともに、経時的に芽の呼吸量、エチレン生成量および含水率を測定した。両時期とも対照区よりもH2CN2処理区の発芽が早く、しかも休眠の深い12月処理で区による差が大きかった。両区いずれの時期とも、芽の呼吸量は発芽直前に急上昇したのに対し、エチレン生成量は調査期間を通して低いままであった。芽の含水率は、いずれの時期および処理区とも発芽期に低下し、特にH2CN2処理区の低下が大きかった。休眠最深期の10月に採取した穂木にACC、GSH(還元型グルタチオン)およびGSSG(酸化型グルタチオン)を処理し、発芽に及ぼす影響を調査したところ、ACCだけが発芽を促進した。同様に、4種のシアン化合物(CaCN2、H2CN2、KCN、NaCN)を処理したところ、H2CN2を除き有意に発芽を促した。これらの結果を基に、ブドウの発芽と生理的変化との関係および休眠覚醒に及ぼすエチレン生合成関連物質の作用性について考察した。

    Using 'Beni Shimizu' peach (Prunus persica (L.) Batsch) trees maintained by light pruning with heavy disbudding (LP) or conventional method (CM) as heavy pruning with no disbudding fruit development was investigated in relation to the occurrence of reddish-pulp for four years. Fruits harvested from LP trees had a higher rate of reddish-pulp development, whereas in fruits harvested from CM trees the percentage was low, irrespective of the year surveyed. LP trees resulted in greater yield compared to CM trees. Nitrogen content in the flesh of fruits was significantly higher in LP trees than in those of CM trees during stage 1 of fruit development. When carbohydrate concentrations in the cane and root of LP and CM trees were compared between before and after full bloom, the former trees showed a greater decrease than the latter. Throughout fruit development, the diameter of cells in the flesh of fruits from LP trees was significantly larger than those from CM trees. Besides, the fruit growth rate was greater in the former than in the latter at stage 3 of fruit development. Fruits harvested from LP trees attained higher weight than those from CM trees. Total soluble solid contents of the juice in fruit harvested were significantly higher in LP trees than in CM trees. In LP trees, shoot growth ceased by the beginning of stage 2 of fruit development, while in CM trees, shoot growth continued until stage 3. In LP trees, anthocyanin concentration in the flesh of fruit increased rapidly just before harvest. Total phenolic concentration in the flesh was higher in fruits from LP trees than in those from CM trees throughout fruit development. Further, there was a higher L-phenylalanine ammonia-lyase (PAL) activity in fruit from LP trees than in those from CM trees. Based on these results, the relationship between pruning levels and the occurrence of reddish-pulp in peach fruit is discussed.<br>

    Seven potted dormant tetraploid grapevine cultivars grown under forced conditions were treated immediately after pruning with the supernatants of a 20% suspension of CaCN2, 2% H2CN2 and 5% diallyl disulfide in different chilling exposures: late November (73 hours), early January (620 hours) and late February (1,275 hours). The effectiveness of the chemicals in initiating budbreak was assessed by the number of days to initial and 60% budbreak after treatment, indicating the promotion and uniformity of budbreak, respectively. Although the chemicals were most effective when applied in November, there was large variation in the response of buds among cultivars and treatment times. For November treatment, H2CN2 was the most effective in accelerating budbreak initiation and enhancing uniformity for almost all cultivars, followed by CaCN2, whereas in the January and February treatments, there were no differences in the promotion and uniformity of budbreak between the two chemicals. Response of buds to diallyl disulfide varied largely among the cultivars. Diallyl disulfide delayed budbreak initiation or reduced percentage budbreak for 'Aki Queen' and 'Takatsuma' in all the treatment times and for 'Pione' in November and January treatments. For 'Fujiminori,' there was no significant difference in the effectiveness of chemicals on budbreak regardless of treatment time. We conclude that H2CN2 is the most effective chemical for budbreak of tetraploid cultivars, followed by CaCN2, and diallyl disulfide. However, the response of grapevine buds to chemicals varies with the depth of dormancy and cultivar.<br>

    We investigated the morphology of seed, particularly of the endosperm of intact and abscising 'Shimizu Hakuto' peach (Prunus persica Batsch) subjected to different fruit thinning levels for four years. The objective was to study the anatomical basis for physiological fruit drop which occurs from end of Stage 2 to beginning of Stage 3 of fruit growth. The percentage of physiological fruit drop was significantly higher (P=0.05) in trees subjected to heavy fruit thinning (HFT) than in those subjected to light fruit thinning (LFT) or standard fruit thinning (SFT). The mean endosperm length in fruit which dropped at the peak of physiological fruit drop from trees under HFT was equal to that of intact fruit collected at 65 days after full bloom (DAFB); the mean embryo length in dropped fruit was shorter than that in the intact fruit collected at the same time. Endosperm and embryo cells in dropped fruit showed typical necrotic symptoms, such as cells with shrunken cytoplasm and collapsing cell nucleus. Collapsing cell nucleus in the endosperm of the intact fruit collected from trees under HFT began at 66 DAFB that corresponded with the time when the endosperm stopped growing. Morphology of chalazal haustorium was observed in the on-tree fruit collected from the beginning to the middle of Growth Stage 2 in trees under HFT and SFT. Although most chalazal haustoria had reached the chalaza at the beginning of Growth Stage 2 in both fruit thinning levels, those in most of HFT fruit exhibited abnormal shrinking at 54 DAFB. These results indicate that a series of events take place in the seeds predestined to drop. That is, after certain factor(s) that cause shrinking of the chalazal haustorium and retardation of embryo growth to occur, assimilated reserves in the endosperm cannot be translocated to the embryo because of shrinkage of the chalazal haustorium. Finally, necrosis of embryo is induced.

    The relationships between fruit enlargement and development of seed components, particularly embryo were investigated in 2000 and 2001 with respect to physiological fruit drop by using 'Shimizu Hakuto' peach (Prunus persica Batsch) trees subjected to different fruit thinning levels. As the standard fruit thinning (SFT), the first thinning was conducted in early May to 2,3-fold levels as final crop load (about 70 leaves per fruit at harvest), followed by the second thinning in early June to the final level. For heavy fruit thinning (HFT), fruit density was adjusted in early May to the final crop load and for light fruit thinning (LFT), in early June to 1.5-fold of the final crop load. The percentage of physiological fruit drop was significantly hie-her (P &lt; 0.05) in trees under HFT than in LFT and SFT in both years. The percentage of split-pit fruit was significantly higher (P &lt; 0.05) in HFT than LFT in 2000 but there was no significant difference between HFT and SFT in 2001. At the end of Growth Stage 2, fruit weight was proportional to seed size, but it was inversely related to embryo size in 2000 under both HFT and LFT. The mean embryo length of HFT fruit was two-thirds of that of LFT, although the different thinning levels had no significant effect on endosperm length. There was no difference in embryo length between those of split-pit and non-split-pit fruit at early Growth Stage 2. However, at the late Growth Stage 2 in HFT trees, the embryos diverted into two types: normal and very small (&lt; 3 mm); this diversion was not observed in SFT. In HFT trees, there was an inverse relationship between fruit weight and dry matter accumulation of seed components. The larger the fruit, the smaller was the dry matter accumulation in the nucellus and endosperm. Thus, the increased rate of physiological fruit drop in HFT trees might be attributed to the reduction of pholosynthates partitioned to the seed because of rapid fruit enlargement which leads to inferior embryo development.

    Five peach (Prunus persica Batsch) cultivars were characterized for the "reddish -pulp" physiological disorder in which the flesh of mature fruit undergoes a pink to red discoloration. Fruit exhibiting this disorder had high contents of anthocyanin and phenolic compounds. The degree of "reddish- pulp" development had a negative significant correlation (P&lt;0.001) with flesh firmness and had no clear relationship with fruit weight and-total soluble solids content. Furthermore, there was no significant difference in the rate of "reddish- pulp" development among cultivars and orchards. However, there was a large difference in the rate of "reddish- pulp" occurrence; with some trees almost 100% of the fruits exhibited the symptom, whereas fruits from other trees expressed a very low percentage, irrespective of the orchard. The degree of "reddish- pulp" in the fruit harvested from trees showing a high rate of discoloration increased after storage at 25 degrees C compared to the fruit harvested from trees showing little or no symptoms of the disorder. The annual difference in both trees, showing high and low rates of "reddish -pulp" occurrence, was small. However, once "reddish- pulp" fruit appeared in a tree, such trees produced fruit with "reddish- pulp" in subsequent years. When the dry weight contents of roots from trees with high and low rates of "reddish -pulp" occurrence were compared in autumn, the former weighed less than the latter probably because it had fewer roots, especially fine roots of less than 2 mm in thickness. Based on these observations the possible cause(s) of the development of "reddish -pulp" in peach fruit is discussed.

    This study characterized the "watercore-like" disorder in which the flesh of mature fruit in peach (Prunus persica Batsch) exhibits a water-soaked brown discoloration. There were varietal differences in the incidence of the disorder with 'Kawanakajima Hakuto, ''Beni Shimizu' and 'Hana Shimizu' exhibiting higher rates. The incidence of disorder in 'Hana Shimizu' fruit ranged from 23 to 54% over the three years. In all cultivars, the disorder was highest on the side opposite to the fruit suture, then both cheeks and the suture, in that order. Fruit with the disorder showed higher weight, and total sugar, sucrose, sorbitol, water soluble pectic substances and total phenolic contents but lower flesh firmness, total amino acid concentrations and hydrochloric acid soluble pectic substances compared to normal fruit. The highest concentration of total soluble solids was observed at the portion of both cheeks of the fruit, followed by the opposite side of the suture and the suture. Symptoms of the disorder were prominent in mature fruit. We propose that "water-soaked brown flesh" be considered a common term for this disorder based on the fact that the symptoms of "watercore-like" disorder as it is commonly known in Okayama were similar to those of "watercore" or "internal browning" disorder, which is the popular term used in other peach producing areas in Japan.

    The possible causes of physiological fruit abscission (June drop) in peach (Prunus persica), which occurs between late Stage 2 and early Stage 3, was investigated for two years. The characteristics of fruit development in 'Shimizuhakuto (SH)' which has a high frequency of pit splitting and physiological fruit drop were compared with those in 'Takeihakuho (TH)' and 'Megohime (MH)' which tend to have numerous split pits but low fruit drop and with 'Yamatohakuto (YH)' which manifest a few split pits acid a small percentage of fruit drop. SH had a lower percentage of fruit drop than YH but its growth rate during Stage 1 was Easter than that of YH. The pits of SH and TH, based on fresh weight and size, grew significantly faster than those of YH throughout the entire developmental period. Seeds of SH elongated faster than those of YH but the former accumulated dry matter slower than did those of the latter after the middle of Stage 2. There was no difference in the dry matter accumulation of the pits between two cultivars. Throughout Stages 2 and 3, the length and cell diameters of embryos were smaller in the fruits of SH than in those of YH. In the former, the sizes of pits and embryos in the split-pit fruits were significantly larger than those in fruits with intact pits. The percentage of dry matter in seeds of split-pit fruits in SH varied widely, but at the heavier end on the distribution, the dry weights were similar in all fruits. Embryo lengths from SH fruits with split-pit was classified into normal and short categories, indicating that some seeds developed normally until pit-splitting stage. Seed weight and the sizes of endosperm and embryo in SH fruit which abscised were significantly smaller compared with those of normal ones, but the weight and size of pits were similar. When fruits of SH trees were thinned to two crop levels, heavy thinning resulted in larger fruits but with a higher percentage of split pits with small embryos and more fruit drop than lightly thinned tree. The high percentage of poorly developed embryos that existed in June drop fruits regardless of thinning level, suggests that inferior seed development prior to pit-splitting is related to June drop in SH.

Both experiments were conducted for 13 weeks beginning at the end of December when the natural daylength corresponded to about 9 hr and 50 min. The primary shoot was pinched to 15 nodes and all lateral shoots were repeatedly pinched, leaving just one leaf at the node. In Exp. 1 an additional lighting was supplied with a homolux lamp as follows: 6 hr after sunset, 6 hr before sunrise, 3 hr after sunset and 3 hr before sunrise, and from sunset to sunrise. The elongation of the primary shoot, the number of leaves on lateral shoots per main shoot and leaf area per shoot were significantly increased when the daylength was extended as compared to the control vines grown under natural daylight. The mean number of bunch primordia per lateral bud in vines exposed to lamp increased three to four- fold compared with the control. No significant difference in shoot growth and flower bud differentiation was observed among the three plots for which daylength was prolonged to 16 hr. Night interruption for 2 hr from 8 p.m., 11 p.m., and 2 a.m, with a homolux lamp (Exp. 2) promoted shoot growth compared to the control vines; the number of bunch primordia was almost the same. There was no significant difference in shoot growth and the flower bud differentiation among the three night break treatments. Among the plots the most vigorous shoot growth and the largest number of bunch primordia per bud occurred in vines exposed to constant (24 hr) light.

    A method for collecting the vacuolar contents of intact tannin and parenchyma cells of persimmon (Diospyros kaki Thunb.) fruit using a micropipette mas developed, Thin sections of the mesocarp tissue from mature persimmon fruit, 'Miyazaki-mukaku' and 'Hiratanenashi', mere placed on a glass slide, Using a micromanipulator and an inverted microscope, a micropipette mas inserted into a vacuole and its contents were withdrawn. A 5-nL sample of vacuole sap was collected per tannin cell from 'Hiratanenashi' and 7 nL from 'Miyazaki-mukaku', whereas only 2 nL was withdrawn from adjacent parenchyma cells, Analyses of the vacuolar sap revealed that the tannin cells of both cultivars contained 10% to 12% (m/v) of tannin as (+)-catechin equivalents and 10% to 13% (m/v) of soluble sugars, whereas the parenchyma cells contained trace amounts of tannins and approximate to 20% of soluble sugars, Tannin cells contain only a slight amount of sucrose, in contrast to a relatively large amount in parenchyma cells.

    In Japanese peach culture, unnecessary flowers (buds) such that attach uprightly or toward centre of tree on the bearing shoots are removed simply and lightly by flower thinning and their fruiting levels are adjusted by fruit thinning done at two steps to control number of fruit remaining on the tree. However, fruit quality still varies widely within a tree and flesh disorders such as "reddish pulp" and "water-soaked brown flesh" occur in many of harvested fruit in recent years. We investigated the relationship between fruiting positions or flowering time and the variation in weight, soluble solids content (SSC) and occurrence of flesh disorders of fruit within a peach cv. white flesh type 'Benishimizu' tree. Fruit weight and SSC were significantly inferior in not only the fruit located on the bottom part of the main stem but also for the fruit borne from early blooming flowers and the rate of fruit showing severe "reddish pulp" disorder was also higher in these fruit. The fruit borne from early blooming flowers had lower sugar accumulation in the flesh than those from late blooming flowers during the latter half of Stage 3 of fruit growth. At present, we are trying to improve flower thinning so as to remove the flowers opening before full bloom. On fruiting shoots of white flesh type 'Shimizu Hakuto' peach that received thinning of early blooming flowers, the distribution rates of fruit that had weights less than 280 g were lower than those of fruiting shoots (= control) that received the standard fruit thinning. Occurrence rate of severe "reddish pulp" disorder when using this flower thinning protocol were significantly lower than the control. Based on the above results, flower and fruit thinning can improve fruit quality by considering not only the within-tree position of fruit but also the flowering time.

    モモ'紅清水'において種子への傷害が誘起する落果と種子発育との関係を検討した。種子への傷害(傷害処理)として果実の縫合線と逆の位置から種皮まで外径2.5mmのドリルを満開後40、50、60、72および80日に挿入した。落果や種子発育と関係するジベレリンとパクロブトラゾールの種子への注入(注入処理)も傷害処理後に行った。傷害処理、注入処理ともに、満開後40-60日処理には処理した果実が全て落下したのに対し、72および80日処理では、いずれも落下しなかった。60日処理では、果実および種子の新鮮重がともに処理3日後に減少し始めた。胚乳と胚の形態を満開後60、72および80日処理間で比較した。胚乳および胚の成長は60日処理では処理3日後に停止したが、72日処理では胚が処理5日後まで成長し続けた。胚乳と胚の細胞核の崩壊は、処理時の胚長が5mm未満であった60日処理では、処理3日後に生じた。一方、処理時の胚長が約8mmであった72日処理では、胚乳における細胞核の崩壊は60日処理と同様の傾向で生じたが、胚の細胞核は崩壊しなかった。60日処理において合点側吸器の形態を観察したところ、吸器は傷害処理や注入処理によって直ちに収縮し始め、正常な合点側吸器を持つ種子の割合は急速に低下した。これらの結果に基づいて、種子への傷害による種子の退化過程ならびに胚の成長と落果との関係を考察した。

    The effects of fruit position in the canopy, harvest date and covering fruit with bags of different colors on the development of reddish-pulp, skin and flesh anthocyanin concentrations and flesh firmness were investigated in 'Takei Hakuho' peach (Prunus persica Batsch) fruit. In the first half of harvesting, the rate of reddish-pulp development was higher in fruit harvested from the upper part of the trees than in those from the lower part, whereas in the last half of harvesting the rate of reddish-pulp development had no relationship with fruit position in the canopy. There was no significant difference in the rate of reddishpulp development among unbagged fruit and those in white and orange bags, However, fruits covered with orange and black bags showed a lower rate of reddish-pulp development at harvest. However, when fruits were stored for four days at room temperature the rate of reddish-pulp development was as high as that in fruit covered with white bags. The relationship between skin and flesh anthocyanin concentrations and flesh firmness was investigated in fruits covered with bags of different colors. There was significant negative correlation (P<0.001) between flesh anthocyanin concentration and flesh firmness and there was no significant correlation observed between skin anthocyanin concentration and flesh firmness. The relationship between skin and flesh anthocyanin concentration in fruit harvested from 'Shimizu Hakuto' peach trees varied largely among the orchards. These findings indicate that anthocyanin accumulation in peach skin is significantly affected by light conditions, such as fruit position in canopy and modification of light quantity through bagging with different colors, although the bags had little or no effect on anthocyanin accumulation in the flesh.

    We investigated the morphology of seed, particularly of the endosperm of intact and abscising 'Shimizu Hakuto' peach (Prunus persica Batsch) subjected to different fruit thinning levels for four years. The objective was to study the anatomical basis for physiological fruit drop which occurs from end of Stage 2 to beginning of Stage 3 of fruit growth. The percentage of physiological fruit drop was significantly higher (P=0.05) in trees subjected to heavy fruit thinning (HFT) than in those subjected to light fruit thinning (LFT) or standard fruit thinning (SFT). The mean endosperm length in fruit which dropped at the peak of physiological fruit drop from trees under HFT was equal to that of intact fruit collected at 65 days after full bloom (DAFB); the mean embryo length in dropped fruit was shorter than that in the intact fruit collected at the same time. Endosperm and embryo cells in dropped fruit showed typical necrotic symptoms, such as cells with shrunken cytoplasm and collapsing cell nucleus. Collapsing cell nucleus in the endosperm of the intact fruit collected from trees under HFT began at 66 DAFB that corresponded with the time when the endosperm stopped growing. Morphology of chalazal haustorium was observed in the on-tree fruit collected from the beginning to the middle of Growth Stage 2 in trees under HFT and SFT. Although most chalazal haustoria had reached the chalaza at the beginning of Growth Stage 2 in both fruit thinning levels, those in most of HFT fruit exhibited abnormal shrinking at 54 DAFB. These results indicate that a series of events take place in the seeds predestined to drop. That is, after certain factor(s) that cause shrinking of the chalazal haustorium and retardation of embryo growth to occur, assimilated reserves in the endosperm cannot be translocated to the embryo because of shrinkage of the chalazal haustorium. Finally, necrosis of embryo is induced.

    The relationships between fruit enlargement and development of seed components, particularly embryo were investigated in 2000 and 2001 with respect to physiological fruit drop by using 'Shimizu Hakuto' peach (Prunus persica Batsch) trees subjected to different fruit thinning levels. As the standard fruit thinning (SFT), the first thinning was conducted in early May to 2,3-fold levels as final crop load (about 70 leaves per fruit at harvest), followed by the second thinning in early June to the final level. For heavy fruit thinning (HFT), fruit density was adjusted in early May to the final crop load and for light fruit thinning (LFT), in early June to 1.5-fold of the final crop load. The percentage of physiological fruit drop was significantly higher (P<0.05) in trees under HFT than in LFT and SFT in both years. The percentage of split-pit fruit was significantly higher (P<0.05) in HFT than LFT in 2000 but there was no significant difference between HFT and SFT in 2001. At the end of Growth Stage 2, fruit weight was proportional to seed size, but it was inversely related to embryo size in 2000 under both HFT and LFT. The mean embryo length of HFT fruit was two-thirds of that of LFT, although the different thinning levels had no significant effect on endosperm length. There was no difference in embryo length between those of split-pit and non-split-pit fruit at early Growth Stage 2. However, at the late Growth Stage 2 in HFT trees, the embryos diverted into two types: normal and very small (<3mm); this diversion was not observed in SFT. In HFT trees, there was an inverse relationship between fruit weight and dry matter accumulation of seed components. The larger the fruit, the smaller was the dry matter accumulation in the nucellus and endosperm. Thus, the increased rate of physiological fruit drop in HFT trees might be attributed to the reduction of photosynthates partitioned to the seed because of rapid fruit enlargement which leads to inferior embryo development.

    Five peach (Prunus persica Batsch) cultivars were characterized for the "reddish-pulp" physiological disorder in which the flesh of mature fruit undergoes a pink to red discoloration. Fruit exhibiting this disorder had high contents of anthocyanin and phenolic compounds. The degree of "reddish-pulp" development had a negative significant correlation (P&lt
    0.001) with flesh firmness and had no clear relationship with fruit weight and total soluble solids content. Furthermore, there was no significant difference in the rate of "reddish-pulp" development among cultivars and orchards. However, there was a large difference in the rate of "reddish-pulp" occurrence
    with some trees almost 100% of the fruits exhibited the symptom, whereas fruits from other trees expressed a very low percentage, irrespective of the orchard. The degree of "reddish-pulp" in the fruit harvested from trees showing a high rate of discoloration increased after storage at 25°C compared to the fruit harvested from trees showing little or no symptoms of the disorder. The annual difference in both trees, showing high and low rates of "reddish-pulp" occurrence, was small. However, once "reddish-pulp" fruit appeared in a tree, such trees produced fruit with "reddish-pulp" in subsequent years. When the dry weight contents of roots from trees with high and low rates of "reddish-pulp" occurrence were compared in autumn, the former weighed less than the latter probably because it had fewer roots, especially fine roots of less than 2 mm in thickness. Based on these observations the possible cause(s) of the development of "reddish-pulp" in peach fruit is discussed.

    モモにおいて音響測定による非破壊判別を簡便に行えるようにするため、モモの核割れ果の判別する試験において、2点型の分析機器「聴診器」と一点型音響装置との間で、共鳴周波数などの音響データの整合性の把握を行った。果実発育中の継続的な変化を確認したところ、おおむね同様の結果となり、1点型試験機でも核割れ発生を判別でき、また果実袋上など直接果実にセンサーを接触させられない栽培状態でも共鳴周波数の取得が可能であることが明らかになった。導入したモバイル型音響装置についても調整を行い、核割れや樹上での成熟状態を把握できるように、プログラムを開発メーカーと調整し、2020年度に果実生産への利用可能性の検討ができるようにした。
    また、一点型音響測定装置を用いて、モモ’白鳳’において、70果実の果実発育中に樹上で共鳴周波数の低下を調査していき、水浸状果肉障害の発生を非破壊把握する方法を検討した。果肉断面の半分程度に水浸症状を呈する水浸指数2以上の商品価値が低下する果実については、正常果と比べて、第2共鳴周波数が著しく低下することから、この閾値を550Hzに設定することで、収穫果において商品性の低い果実の混入を抑制できることが明らかになった。さらに、樹上調査において、7月初めの段階で、ほぼ同じ着果位置でも、共鳴周波数の低い果実が存在し、そのような果実の多くが収穫時に水浸症状をとなっていることが確認された。この方法を利用することで、水浸果となっていく変化が生じ始める時期や着生させている枝の状況などとの関連を検討できることが明らかとなった。これらをまとめて、令和2年度園芸学会春季大会で発表した。

    In order to elucidate the mechanisms regulating ultra-late maturing trait in ‘Gento’ peach, its inheritance in F1 progeny as well as physiological and histological changes during fruit development were investigated. Although F1 individuals that matured as late as ‘Gento’ were not observed, maturity date in F1 progeny exhibited a binominal distribution and 9 bp indel in NAC gene on LG4 co-segregated with the maturity date of all the individuals. Cell size and ABA content in the flesh of ‘Gento’ increased much later than common peach cultivar, corresponding to their fruit growth patterns. These results, combined with transcriptome analysis by RNA-seq and future genetic analysis using F2 progeny, will help us to fully understand the molecular basis for the ultra-late maturing trait in ‘Gento’.

    本年度は、研究グループが注目している棚持ち性の優れる品種、障害発生の異なる枝変わり系統に関して、以下のような成果が得られた。
    1)収穫時期や栽培地域を問わず、‘桃水’では自己触媒的エチレン生合成能が欠損していること、外生エチレンに応答して果実が軟化することが明らかとなり、‘桃水’は硬肉モモに属することが示された。Tatsukiら(2018)による、エチレン生成能の欠損とIAAの生合成関連遺伝子へのトランスポゾンの挿入変異との関連についての報告を参考に、親である‘川中島白桃’および‘桃水’の挿入変異を解析したところ、‘川中島白桃’はヘテロで‘桃水’はホモにて変異を持つことが明らかとなった。さらに、関連する数品種の全ゲノム配列を予備的に調査したところ挿入変異をヘテロで持つ‘ゆうそら’が片親である可能性が示唆された。‘桃水’は外生エチレン処理時の肉質が優れており、韓国系の品種を用いず、日本にて育成された品種同士の交配による高品質かつ棚持ち性の良い品種育成が実証された。
    2)‘大寿蜜桃’ は収穫時期、栽培地域によらず、自己触媒的エチレン生成能を持つにも関わらず、エチレンによって軟化せず、3週間以上の棚持ち性をした。一方、‘冬美白’ではエチレン生成とともに、果肉軟化が進行し、通常のモモと同様のエチレン応答性を示した。
    3)‘紅博桃’およびその亜主枝単位の枝変わりについて調査したところ、枝変わりでは生長第2期(肥大停止期)から3期(最終肥大期)への転換が遅れており、熟期が3週間ほど遅れことや収穫後に蜜症障害が多発することが明らかとなった。
    4)上記の1)-3)の品種や系統に関して、予備的な全ゲノムシークエンス解析を実施し、‘桃水’と‘川中島白桃’の親子関係や、‘紅博桃’樹内の熟期が異なる枝の枝変わり関係を確認するとともに、詳細な解析に向けた方向性を確認した。

    Authors had been studying to know water content in leaves as the biological information, so far we have developed ‘water stress indicator sheet’ using cobalt chloride for simple estimation of water status (Morinaga et.al. 2012, 2016). Now, we are trying to understand water contents in leaves as electrical method with plants.
    Electrostatic capacitance and electro-cycle length were measured using an instrument that we developed, and checked the relationship with transpiration rate by leaf porometer. As a result, the electrostatic capacitance has not clear correlation with transpiration rate because it is too sensitive against electronic noise such as contact, oscillation or length of electronic cord. On the contrary, electro-cycle length had stably good relationship with transpiration rate. It is possible to estimate water status of plants leaves by measuring electro-cycle length.

    Under white and UV fluorescent light, anthocyanins in Gros Colman gapes started to accumulate 48 hr after, illumination. After 72 hr, anthocyanin concentration was markedly increased by white light. Under UV lighting, a considerable amount of anthocyanins also accumulated relative to dark conditions. As white and UV light intensities increased, anthocyanins gradually increased, but promoting effect of light on anthocyanin accumulation was saturated at 60 μmol・m^<-2>・S^<-1> for white and 0.4 W・m^<-2> for UV. Using LEDs emitting specific wavelengths, the action spectrum for anthocyanin accumulation was determined. Among the visible light wavelengths, blue light largely enhanced anthocyanin accumulation, while the green to yellow range was less effective.
    The effects of UV-A and ABA treatments on berry coloration were compared among eight grape cultivars, the red type (Aki Queen, Ruby Okuyama, Hiro Humburg and Kaiji) and the black type (Campbell Early, Pione, Muscat Bailey A and Gros Colman). In Gros Colman, Pione and Aki Quart grapes, the highest level of anthocyanin was observed in clusters treated with ABA, followed by UV-A and then controls, whereas in Hiro Humburg it was high in UV-A than ABA treatment. For Ruby Okuyama, anthocyanin level in clusters treated with ABA and UV-A was almost same. High content of anthocyanin resulted in high activity of PAL in the skin.
    Three cDNA clones, encoding phenylalanine ammonia-lyase were isolated from grape (Vitis labruscana, cv Pione) skin by RT-PCR and RACE-PCR methods. A strong expression of the three genes and a high PAL activity was detected at early stage of fruit growth, followed by a reduction of the expression and activity to trace level at veraison. While in white type cultivars such as Muscat of Alexandria gene expression and enzyme activity remained at trace level during fruit ripening, in black type cultivar such as Pione and Gros Colman the increase in gene expression and enzyme activity during ripening stage were observed, correlating with anthecyanin synthesis. The increase in gene expression and enzyme activity were dramatically stimulated by ABA treatment at veraison.

    The total phenolic contents in the mature 'Hakuto' peach fruits varied widely among the orchards while significant difference among trees in the same orchard was not observed. Phenolic content ded not correlate with the contents of tatal solublee solids, fruit weight, and tree age. The larger the total phenolic contents, the higher was the EC of soil, but the relationship between total phenolic contents and soil pH was not observed.
    Regardless of orchard, phenolic contents in fruit decreased towards the end of Stage 1 of fruit growth, after which it increased rapidly and reached a maximum in the middle of Stage 2. Phenolic contents rapidly decreased to a lower level during Stage 3. Throughout fruit growth, total phenolic contents were larger in the fruits from orchard which phenolic content in the mature fruits was high than in the fruits from orchard which it was low, especially during Stage 2. The relationships between phenolic contents and the shoot growth and soil moisture were not observed. PAL (L-phenylalanine ammonialyase) activity was high during Stage 1 and reached a maximum in the middle of Stage 2, after which no activity detected. Throughout Stage 1 and the first half of Stage 2, the larger the phenolic contents, the higher was the PAL activitiy in fruit. However, no relationship was observed between the PAL activity and the contents of N, Mn, Cu and B which have been related to phenolic metabolism.
    Total phenolic contents in the mature fruits were the highest in Mishima-hakuto and Golden Peach, followed in Okubo and Hakuto, while it was very low in Kawanakashima-hakuto and nectalin. In almost cultivars, molecular weight of phenolics was large in Stage 1 and was low in Stage 2, after which it increased again during Stage 3. The HPLC analysis showed that six phenolic compounds were present in peach fruit irrespective of cultivar. Content of catechin was larger than in that of chlorogenic acid during Stage 1 and 2, while in stage 3 latter was larger than in the former.
