Internal detoxification mechanism of Al in hydrangea - Identification of Al form in the leaves

An internal detoxification mechanism for Al was investigated in an Al-accumulating plant, hydrangea (Hydrangea macrophylla), focusing on Al forms present in the cells. The leaves of hydrangea contained as much as 15.7 mmol Al kg(-1) fresh weight, and more than two-thirds of the Al was found in the cell sap. Using Al-27-nuclear magnetic resonance, the dominant peak of Al was observed at a chemical shift of 11 to 12 parts per million in both intact leaves and the extracted cell sap, which is in good accordance with the chemical shift for the 1:1 Al-citrate complex. Purification of cell sap by molecular sieve chromatography (Sephadex G-10) combined with ion-exclusion chromatography indicated that Al in fractions with the same retention time as citric acid contributed to the observed Al-27 peak in the intact leaves. The molar ratio of Al to citric acid in the crude and purified cell sap approximated 1. The structure of the ligand chelated with Al was identified to be citric acid. Bioassay experiments showed that the purified Al complex from the cell sap did not inhibit root elongation of corn (Zea mays L.) and the viability of cells on the root tip surface was also not affected. These observations indicate that Al is bound to citric acid in the cells of hydrangea leaves.

Two inbreds of Zea mays, tolerant to K^+ deficiency and control, were grown in the presence of different concentrations of K^+. Several parameters were compared between the two inbreds and characteristic differences in terms of K^+ utilization were observed. 1. The dry weight of both inbreds was nearly the same in the presence of a sufficient concentration of K^+, but was apparently higher in the tolerant inbred than the control inbred according to the decreasing amount of K^+ supply. 2. Symptoms of K^+ deficiency in the tolerant inbred appeared only with complete absence of K^+, but those in the control inbred could be observed even at a decreased level of K^+ supply. 3. The increased length of the main root and top to root ratio in the control inbred were conspicuously lower than those of the tolerant inbred due to K^+ deficiency. Furthermore, the amout of lateral roots of the cotrol inbred was approximately one-half to one-fourth of that of the tolerant inbred. 4. The capability of K^+ absorption by the roots treated for K^+ deficiency was higher in the tolerant inbred than in the controel inbred. The difference in the capability became larger when assayed with decreasing concentrations of K^+. 5. K^+-dependent H^+ extrusion from roots treated for K^+ deficiency was larger in the tolerant inbred than in the control inbred.

Effective regulation of iron acquisition in graminaceous plants. The role of mugineic acids as phytosiderophores

Discovery of mugineic acids as phytosiderophores has shown that some graminaceous monocotyledonous plants have a different iron acquisition strategy (strategy II) from dicotyledonous and nongraminaceous monocotyledonous plants (strategy I). The process of iron acquisition by strategy II plants can be divided into four main steps: biosynthesis, secretion, solubilization, and uptake, all of which are effectively regulated by different systems. The biosynthesis of mugineic acids is controlled by an on-off system which is operated under the control of iron demand in the plant. All mugineic acids share the same biosynthetic pathway from L-methionine to 2'-deoxymugineic acid, but the subsequent steps differ among plant species and even cultivars. The biosynthesis of mugineic acids is associated with the methionine recycling pathway. The secretion of mugineic acids shows a distinct diurnal rhythm. Mugineic acids solubilize sparingly soluble inorganic iron by chelation and possess a high chelation affinity for iron, but not for other polyvalent ions such as Ca2+, Mg2+ and Al3+. The iron uptake process is regulated by a specific uptake system that transports the mugineic acid-Fe(III) complex as an intact molecule. This system specifically recognizes the mugineic acid-Fe(III) complexes, but not other mugineic acid-metal or synthetic chelator-Fe(III) complexes, suggesting that binding sites with strict recognition for stereostructure of the complex are located on the plasma membrane. All these regulatory systems are considered to represent an efficient strategy to acquire adequate amounts of iron and to avoid factors unfavorable for iron acquisition such as high pH, high concentrations of bicarbonate, Ca2+ and Mg2+, microbial degradation, and uptake of other metals that are common in calcareous soils.

Mugineic acids are a group of compounds having ferric chelating activity. They are secreted from the roots of some gramineous plants under iron deficiency. These compounds play an important role in the acquisition of insoluble iron from soils by the gramineous plants. In the present study, the biosynthetic pathways of five mugineic acids were investigated in different gramineous plants using feeding experiments with ^<13>-and/or ^2H-labeled compounds and by NMR techniques. The results revealed that L-Met serves as a precursor for all mugineic acids tested. All mugineic acids share the pathway from L-Met to 2'-deoxymugineic acid although subsequent steps differ and are dependent on the plant species and cultivars. In oats, avenic acid A is biosynthesized from 2'-deoxymugineic acid by cleavage of the azetidine ring. In barley, hydroxylation at the C-2' position in 2'-deoxymugineic acid yields mugineic acid. Further hydroxylation at the C-3 position in mugineic acid produces 3-epihydroxymugineic acid and hydroxymugineic acid in beer barley and rye, respectively. All mugineic acids were found to have similar effect on iron uptake, suggesting that 2'-deoxymugineic acid is a key phytosiderophore. The biosynthetic pathway between L-methionine and 2'-deoxymugineic acid was investigated by observing the incorporation of label from ^<13>C, ^2H, and ^<15>N-labeled methionine in wheat roots (Triticum aestivum L. cv Minori). As a result, only the deuterium atom from the C-2 position of L-methionine was lost, while other atoms were completely incorporated when three molecules of methionine were converted to 2'-deoxymugineic acid. These observations are consistent with the conversion of L-methionine to azetidine-2-carboxylic acid, suggesting that L-methionine is first converted to azetidine-2-carboxylic acid during biosynthesis leading to 2'-deoxymugineic acid. Based on these results, a hypothetical pathway from L-methionine to 2'-deoxymugineic acid was postulated.

The biosynthetic pathway of 2′-deoxymugineic acid, a key phytosiderophore, was investigated by feeding 13C-, 2H-, and 15N-labeled methionine, the first precursor, to the roots of hydroponically cultured wheat (Triticum aestivum L. cv Minori). The incorporation of label from each methionine species was observed during their conversion to 2′-deoxymugineic acid, using 2H-, 15N-, and 13C-nuclear magnetic resonance (NMR). L-[1-13C]Methionine (99% 13C) was efficiently incorporated, resulting in 13C enrichment of the three carboxyl groups of 2′-deoxymugineic acid. Use of D,L-[15N]methionine (95% 15N) resulted in 15N enrichment of 2′-deoxymugineic acid at the azetidine ring nitrogen and the secondary amino nitrogen. When D,L-[2,3,3-2H3-S-methyl-2H 3]methionine (98.2% 2H) was fed to the roots, 2H-NMR results indicated that only six deuterium atoms were incorporated, and that the deuterium atom from the C-2 position of each methionine was almost completely lost. [2,2,3,3-2H4]1-Aminocyclopropane-1-carboxylic acid (98% 2H) was not incorporated into 2′-deoxymugineic acid. These data and our previous findings demonstrated that only the deuterium atom from the C-2 position of L-methionine was lost, and that other atoms were completely incorporated when three molecules of methionine were converted to 2′-deoxymugineic acid. These observations are consistent with the conversion of L-methionine to azetidine-2-carboxylic acid, suggesting that L-methionine is first converted to azetidine-2-carboxylic acid during biosynthesis leading to 2′-deoxymugineic acid. Based on these results, a hypothetical pathway from L-methionine to 2′-deoxymugineic acid was postulated.

Biosynthetic pathways of 3-epihydroxymugineic acid and 3-hydroxymugineic acid in gramineous plants

To elucidate the biosynthetic pathways of 3-epihydroxymugineic acid and 3-hydroxymugineic acid, feeding experiments using 2H- and/or 13C-labeled compounds were conducted with barley (Hordeum vulgare L. cv Ehimehadaka) and rye (Secale cereale L. cv Elubon), respectively. Feeding of [1, 4′, 4″13C3]2′-deoxymugineic acid (25 atom% 13C) to iron-deficient barley yielded mugineic acid and 3-epihydroxymugineic acid enriched with 13C at their corresponding positions. These results enabled us to demonstrate that 2′-deoxymugineic acid acts as a precursor for mugineic acid and 3-epihydroxymugineic acid. The 2H-labeled 3-hydroxymugineic acid was obtained by feeding d, l- [3, 3, 4, 42H4] methionine (98.6 atom% 2H) to iron-deficient rye plants. The 2H-NMR study revealed that two deuterium atoms (one each at the C-2′ and C-3 positions) from the three 2H-labeled methionine molecules were lost in 3-hydroxymugineic acid, whereas the other 10 deuterium atoms were incorporated in a manner similar to that of 2′-deoxymugineic acid and mugineic acid. These findings suggested that 3-hydroxymugineic acid, instead of being biosynthesized from 3-epihydroxymugineic acid by epimerization, was in fact derived from mugineic acid by hydroxylation at the C-3 position. This assumption was further confirmed by the results of the incorporation of 13C-labeled 2’-deoxymugineic acid into 3-hydroxymugineic acid. These results revealed that the biosynthetic pathways of both 3-epihydroxymugineic acid and 3-hydroxymugineic acid are as follows: L-methionineℲ2′-deoxymugineic acid→mugineic acid→3-epihydroxymugineic acid in barley and→hydroxymugineic acid in rye. © Taylor & Francis Group, LLC.

Inhibition of mugineic acid‐ferric complex uptake in barley by copper, zinc and cobalt

The interfering effects of copper, zinc, and cobalt on the uptake of mugineic acid‐ferric complex were studied in barley (Hordeum vulgare, cv. Minorimugi) grown in nutrient solution. Short‐term uptake experiments of 3 h were performed utilizing both ionic and mugineic acid‐complex forms of each metal at two different concentrations. Copper was most effective in decreasing iron uptake when added in an ionic form at either concentration. The inhibition order at higher concentrations followed Cu(II) > Zn(II) ≥ Co(II), Co(III), which is consistent with the stability constants of these metal complexes with mugineic acid. The displacement of iron from its mugineic acid complex by these metals is suggested as a probable explanation for the decreased iron uptake. The inhibitory effect of metal complexes with mugineic acid on iron uptake was only found in cases with higher concentrations of Cu(II) and Zn(II) complexes. Deformation of the specific iron transport system in the plasma membrane due to their adsorption may be responsible for this effect. Copyright © 1993, Wiley Blackwell. All rights reserved

The biosynthesis of mugineic acids was studied by feeding 2H- or 13C-labeled compounds to water-cultured roots in several gramineous plants. The fate of labeled compounds was monitored by using 2H- and 13C-nuclear magnetic resonance. On investigating the proton changes during biosynthesis by feeding D,L-[3,3,4,4-d4]- methionine (98.6% 2H), 2H-labeled 2′-deoxymugineic, mugineic, and 3-epihydroxymugineic acids were isolated from root washings of wheat (Triticum aestivum L. cv Minori), barley (Hordeum vulgare L. cv Minorimugi), and beer barley (Hordeum vulgare L. cv AM Nijo Tochigi), respectively. The 2H-nuclear magnetic resonance study indicated that 12 deuteriums were incorporated into the labeled 2′-deoxymugineic acid, suggesting that three molecules of L-[3,3,4,4-d4]methionme were combined. In comparison, one of the deuteriums at C-2′ position in the mugineic acid, and one each of the deuteriums at C-2′ and C-3 positions in the 3-epihydroxymugineic acid, were lost. However, all other deuteriums were incorporated in a manner similar to that of the labeled 2′-deoxymugineic acid. When [1,4′,4″-13C3]2′-deoxymugineicacid (20% 13C) was fed to oat roots (Avena saliva L. cv Amuri II), avenic acid A, which was 13C enriched at the corresponding positions, was obtained. These results revealed that L-methionine was the precursor for all these mugineic acids and that cleavage of the azetidine ring or hydroxylation of the 2′-deoxymugineic acid produced two related biosynthetic pathways in different gramineous plant species: L-methionine → 2′-deoxymugineic acid → avenic acid A in oat; and L-methionine → 2′-deoxymugineic acid → mugineic acid → 3-epihydroxymugineic acid in barley and beer barley.

Interaction between calcium and silicon in water-cultured rice plants

The interaction between Ca and Si in water-cultured rice plants (Oryza sativa L. cv. Akebono) was investigated in terms of uptake. The effect of Ca levels in the solution on Si chemical forms and on the formation of silica bodies in the leaf blades was also examined using soft X-ray irradiation for detection of silica bodies. Si addition (1.66 m M Si) decreased both Ca content of the shoot and uptake at each Ca level. This might mainly result from a decreased transpiration rate caused by Si. Si uptake was not affected when the Ca levels were increased. The results of Si forms showed that silica sol constituted more than 90% of the total Si in the leaf blades regardless of Si and Ca levels, and soluble silica and/or polysilicic acid seems to gel physically over 8.0 m M Si within the plants. Significant difference in the numbers of silica bodies on the third leaf blade was not found between different Ca levels at the same Si level. The biosynthesis of avenic acid A was studied by feeding L-methionine-l-13C or D,L-methionine-3, 3, 4, 4-d4 to iron-deficient oat roots (Avena sativa L. cv Amuri II). Both avenic acid A and 2’-deoxymugineic acid labeled with 13C or 2H were isolated from root-washings of the same plants. The 13C-NMR elucidated that three molecules of L-methionine were incorporated into avenic acid A in the same way as 2’-deoxymugineic acid. The 2H-NMR spectra indicated that all deuteriums from the methionine were incorporated into 2’-deoxymugineic acid, but one of the deuteriums at C-4 was lost in avenic acid A. Effect of silicate on phosphate availability for rice in a P-deficient soil

In a pot experiment the effect of silicate on P availability for rice grown in a P-deficient soil with and without flooding was analyzed. Treatments were designed as follows: C (control: Yakuno soil), SS (sodium silicate application, at 0.47 mg Si g-1 soil) and SC (sodium carbonate application). In order to separate pH effect from Si effect, SC was adjusted to the same pH as SS. Soil pH of SS and SC increased by 1.0 unit. Shoot dry weight of SC plants, and more so of SS plants, increased under both nonflooded and flooded conditions. P concentrations in the shoots were not increased under either condition of SS and SC. With SS, Si concentration in the shoots significantly increased, Mn concentration significantly decreased, resulting in a higher P/Mn ratio in the shoot, but not with SC. Both SS and SC increased N concentration in the shoots nearly two times compared with control under both conditions. Adsorption experiments showed that neither SS nor SC decreased P adsorption by soil. SS also could not displace the adsorbed P in soil samples which had previously either received P or not. These results suggest that the beneficial effects of silicate on rice growth do not result from increasing P availability in soil. The Si effect may be attributed to decreasing Mn uptake, thus indirectly improving P utilization in the plant. © 1991 Kluwer Academic Publishers.

Availability of rice straw Si to rice plants

A pot experiment was conducted to estimate the availability to rice plants of Si contained in rice straw. To separate the effect of straw application on Si availability in soil, two kinds of rice straws, which contained 15.5% (+ Si straw) and 0.2% SiO2 (−Si straw), were used at a rate of 1% of air-dry soil, respectively. Rice plants were cultivated twice during a period of 2 months each. Results of the two harvests indicated that +Si straw application increased the Si concentration in the percolating water, the Si content of rice shoots, and soil-available silicate. However, compared to the Si contained in the added straw, the release of Si from the +Si straw was only about 6% in the first cultivation, and 3% in the second cultivation, respectively. The −Si straw application was not associated with any of the effects observed in the +Si straw application. Successive water extractions showed that the solubility of Si in the +Si straw was lower than that of silica gel, but much higher than that of opal. These facts suggest that the short term availability of Si in the straw applied to the soil is low and the application of materials with a higher availability of Si such as slags may be desirable for satisfactory growth of rice plant. © 1991 Taylor & Francis Group, LLC.

The effect of silicic acid on rice in a P-deficient soil

A pot experiment was conducted to analyze the effect of silicon on the growth of rice grown in a P-deficient soil and on P availability in the soil. Silicic acid was used, rather than a silicate salt, to avoid the complication of changes in soil pH. Shoot dry weight on silicic acid treated soil (0.47 mg Si g-1 soil) increased significantly under both nonflooded and flooded conditions. Shoot Si concentration also increased although P concentration did not. Mn concentration decreased with silicic acid, resulting in a higher P/Mn ratio in shoots. An adsorption and desorption experiment showed that silicic acid did not displace P nor decrease the ability of the soil to adsorb P. In contrast, Si desorption increased with increasing P concentration in the solution, and Si adsorption was reduced when P was applied first. These results suggest that silicic acid does not increase P availability in soil. Increased dry weight may be attributed to a higher P/Mn ratio in the shoot, which may improve P utilization in the plant. © 1990 Kluwer Academic Publishers.
