    Abstract

    The oral microbiota plays a vital role in human health, yet most studies have focused on individual bacterial taxa. To provide a more comprehensive understanding, we analyzed tongue microbiota data from 729 Japanese individuals and classified samples into three types (orotypes): Neisseria -dominant (N), Prevotella -dominant (P), and Streptococcus -dominant (S) types. Each orotype exhibited distinct co-occurrence network structures and was associated with lifestyle factors such as oral care, diet, and smoking. The S type was associated with higher odds of abnormal oral health and metabolic syndrome-related outcomes compared to the N type. In addition, we developed a robust classification model (ROC–AUC > 0.95) to predict orotypes, which showed temporal stability in nearly half of individuals over a 6-year period. These findings highlight the value of orotype classification for monitoring tongue microbial communities and suggest its potential in health risk assessment.

    ABSTRACT

    Background

    Tongue–lip motor function is essential for speech and swallowing. However, research on oral diadochokinesis (ODK), an indicator of this function, remains limited among Japanese children.

    Aim

    To provide foundational ODK data in Japanese children and examine its associations with grade, sex and dental and anthropometric variables.

    Design

    Data from the 2022–2023 Kuroishi Oral Health Study were analysed for 464 third‐ to sixth‐grade students. ODK (/pa/, /ta/, /ka/), grade, sex, total number of teeth, decayed, missing and filled permanent teeth (DMFT) index, and body mass index (BMI) were examined using analysis of variance, t ‐tests, and multiple regression.

    Results

    Mean ODK values increased with grade, with sixth‐graders showing significantly higher values than third‐, fourth‐ or fifth‐graders. Across all syllables, girls showed significantly higher values than boys in fourth or fifth grades, with no significant differences in third or sixth grades. No significant associations were found between ODK and the total number of teeth, DMFT index or BMI.

    Conclusion

    This study provides foundational ODK data in Japanese children, highlighting grade‐ and sex‐related variations. These findings enhance the understanding of tongue–lip motor function development and may serve as a reference for future research and clinical assessments.

    Background/Objectives: The urine sodium/potassium (Na/K) ratio can potentially be used to detect dietary habits that contribute to hypertension. In this prospective cohort interventional trial, we aimed to verify whether private insurance sales staff can help clients change their lifestyle habits based on their urinalysis results. Methods: Clients of the life insurance company (20–65 years old) who were considered to have “high risk” lifestyle factors, which was defined as having high values for two or more of the following indicators: body mass index, blood pressure, triglycerides, liver enzymes, and glucose metabolism, were included. The clients were randomly assigned to three groups: a face-to-face (FF) intervention by sales staff (n = 83), non-FF (Non-FF) intervention via a social networking service (n = 87), and no intervention (Control) (n = 58). Urinalysis and surveys about diet and exercise habits were conducted before and after a 3-month interventional period in all groups. Three interventions were performed for the FF and Non-FF groups, including dietary advice based on urinalysis results, education encouraging reduced salt intake and increased locomotor activity, and viewing an educational video. The Control group only received their urinalysis results by mail. Results: The participants’ mean age was 44.0 years old. Significant improvements in estimated potassium intake were observed in the Non-FF group, and significant reductions in urine Na/K ratios were noted in both the FF and Non-FF groups. Multiple logistic regression analysis indicated that watching the video was the most effective factor for decreasing the urine Na/K ratio (odds ratio = 1.869). The total points for dietary behavior, based on the questionnaire, significantly improved among the individuals who watched the video. Conclusions: This study demonstrates the potential for private health insurance companies to contribute to health promotion and introduces a novel strategy for improving lifestyle habits among individuals at high risk of lifestyle-related diseases.
