    PURPOSE: The lack of occlusal support is an epidemiological risk factor linked to Alzheimer's disease. This study sought to assess the relationship between amyloid β (Aβ) deposition and the lack of occlusal support in amyloid precursor protein (APP) knock-in mice. METHODS: Sixteen experimental animals were divided into two groups. The upper molars were extracted in the extraction group (group E), and a sham operation was performed in the control group (group C). The Morris water maze test was performed 4 months after the tooth extraction. Aβ immunohistochemical staining and Nissl staining of the hippocampus were performed. Hippocampal plasma corticosterone and Aβ protein levels were measured. RESULTS: In the maze task, the escape latency was significantly longer in group E than in group C. In the probe trials, the time elapsed in the target quadrant was significantly shorter in group E than in group C. The number of hippocampal neurons decreased in group E. There was no significant difference in the plasma corticosterone levels between the two groups, indicating that there was no effect of chronic stress on the behavioral results. Hippocampal Aβ40 and Aβ42 protein levels and Aβ deposition areas by immunohistochemical staining were not significantly different between the two groups. CONCLUSIONS: Aβ deposition was not increased in the hippocampus of molarless APP knock-in mice. As such, it appears that cognitive impairment due to a lack of occlusal support was not related to Aβ deposition.

    PURPOSE: Animal experiments have shown that the loss of occlusal support causes impairments in spatial cognition. Many reports have focused on the memory encoding process, and only few studies have investigated the effect on memory retrieval. This study aimed to examine the effects of both the loss and reconstruction of occlusal support on the memory retrieval process and on the number of hippocampal pyramidal cells. METHODS: The experimental animals were divided into a molarless group, in which molars were extracted, a denture-wearing molarless group with experimental dentures inserted after molar extraction, and a control group. Radial maze trials were performed for 20 consecutive days (acquisition phase). The rats were tested on long-term memory retention following the acquisition phase in eight trials every five days, and in an additional trial 20 days later (probe phase). RESULTS: The denture-wearing molarless group showed higher levels of spatial learning and memory than the molarless group. There were no significant differences in memory retrieval during the probe phase between the denture-wearing molarless and the control group. The molarless group showed significantly worse spatial learning and memory and had fewer neurons in the hippocampus than the control group. CONCLUSIONS: Our results suggest that the loss of occlusal support decreases the number of pyramidal cells in the hippocampus and impairs memory decoding and retrieval. However, this effect is suppressed by the reconstruction of occlusal support.

    OBJECTIVES: The present study aimed to identify the effect of memory-related genes on male rats tested for spatial memory with either molar teeth extraction or its restoration by occlusal support using experimental dentures. DESIGN: Memory-related genes were detected from hippocampi of male Wistar rats (exposed to teeth extraction with or without dentures, or no extraction (control)) (7-week old) after behavioural testing (via the radial maze task) using a DNA microarray. The time course of the expression of these genes was evaluated by quantitative real-time polymerase chain reaction (PCR) (on 49-week-old rats). RESULTS: In preliminary experiments, to determine which memory genes are affected by spatial memory training, DNA microarray analysis revealed that thyrotropin-releasing hormone (Trh) and tenascin XA (Tnxa) were up-regulated and neuronatin (Nnat) and S100a9 were down-regulated after the maze training. The expression of Tnxa, Nnat and S100a9 of 49-week-old rats (during the time course) via quantitative real-time PCR was consistent with the results of microarrays of the preliminary experiment. Expression of Trh that was evaluated by quantitative real-time PCR did not agree with the results for this gene from the microarray for all groups. Therefore, expression of Trh may have increased in only young, trained rats. The expression of S100a9 prior to the maze task was down-regulated in only the extraction group. CONCLUSION: These results demonstrated that Trh, Tnxa and Nnat genes were affected according to the degree of memory in male rats. This study also indicated that S100a9 is a memory-related gene, which is affected by the presence of occlusal support.

    OBJECTIVE: Intensive mechanical stress and/or inflammation are known to induce alveolar bone resorption. This study investigated whether a distribution of mechanical stress would reduce residual ridge resorption or improve ischaemia. DESIGN: Thirty rats were divided into six experimental groups (n=5). The control group received no intentional stimulation, but rats in the experimental groups wore denture stimulators made of acrylic resin or a soft lining material. The stimulator transmitted masticatory pressure to the rats' palates for four weeks. The four types of soft lining materials investigated in this study dispersed the applied pressure, with compressive stress ranging from 20.8 to 90.8kPa. Volumes of blood flow and bone resorption of denture foundations were measured every week for 4 weeks. Statistical evaluation of these results was performed using two-way ANOVA and Holm-Sidak test within 5% error limits. RESULTS: Non-viscoelastic material clearly induced bone resorption and ischaemia of denture foundations, while viscoelastic materials reduced these phenomena to different extents according to their viscoelastic properties. Ischaemia in the alveolar ridge preceded residual ridge resorption, because the amount of residual ridge resorption and blood flow rate showed a simple linear regression. CONCLUSION: Animal model of this study suggested that a distribution or reduction of mechanical stress could improve blood flow and decrease alveolar ridge resorption.

    Background: Expression of argyrophilic nucleolar organizer regions (AgNOR) was counted on mucosal epithelium to detect whether the AgNOR count (the number of AgNOR per nucleus) is, for 20 weeks, affected by streptozotocin-induced diabetes mellitus.
    Methods: Diabetic rats were applied non-denture wearing, non-pressure contact, continuous pressure (0.5, 1.5 or 3.4 kPa) and intermittent pressure (10.5, 33.0 or 81.7 kPa), respectively.
    Results: In the non-denture wearing or the non-pressure contact group, AgNOR count was almost constant (ranging 1.86-1.92) without significant difference. Under continuous pressure, AgNOR count significantly decreased at earlier stage and finally recovered to the non-pressure contact group. A similar effect of continuous pressure appeared to manifest itself in intermittent pressure, but to a greater degree. Consequently, AgNOR count under intermittent pressure showed no recovery at 20-week stage.
    Conclusion: Diabetes mellitus exerted, by itself, no influence on the cellular proliferative activity under continuous pressure, whereas the activity deteriorated under intermittent pressure.

    In this study, the experimental denture base on the hard palate of the molar region of osteoporotic rats was designed to cover without any mechanical pressure or to load continuous pressure (0.5, 1.5 or 3.4 kPa). Experimental osteoporosis was induced by ovariectomy and feeding on a low calcium diet. Histopathological changes induced by non-pressure covering or continuous pressure in the tissues under the experimental denture bases were examined. Regarding the histological changes induced by continuous pressure, this was evaluated in relation to the time course of the intensity of the continuous pressure. In the tissues under the denture base in osteoporotic condition with non-pressure covering, no histopathological changes caused by non-pressure covering were observed except for a transient slight proliferative reaction in the epithelial ridge. The following histopathological changes were observed in the tissues under the denture base in the osteoporotic condition bearing continuous pressure; (i) bone resorption was induced on the periosteal surface (Ps) in the osteoporotic-3.4 kPa group, and on the pressure side of the endocortical surface (p-Ec) in the osteoporotic-1.5 and -3.4 kPa group, (ii) the appearance of osteoclasts on the p-Ec in the osteoporotic-3.4 kPa group was earlier than that on the Ps in a same group, (iii) bone resorption on the pressure side of the Ec in the osteoporotic-3.4 kPa group manifested at the earlier stage than that in the osteoporotic-1.5 kPa group, (iv) bone formation following bone resorption on the p-Ec was observed more markedly than that on the Ps and (v) histopathological changes in the tissues under the denture base in experimental osteoporosis bearing continuous pressure had an intimate relationship with the initial intensity and the time course of continuous pressure.

    The effect of calcium supplementation on the bone dynamics in the hard palate of the molar region (maxilla), mandible and proximal tibia in experimental osteoporotic rats was examined. Ninety ovariectomized (OVX) and 45 sham-OVX Wistar female rats were used in this study. All the rats received surgical operation at 6 weeks of age. Ovariectomized rats were fed on a low calcium diet (0.02%) for 12 weeks post-operation, and then randomly divided into the two following groups. One group was fed on high calcium diet (2.30%) (OVX-HCa) and the other group was remained on the low calcium diet (OVX-LCa). Sham-OVX rats were fed on regular calcium diet (1.15%) during the experimental period (Sham-OVX). Histomorphological analysis was carried out from 12 to 32 weeks postoperation. On undecalcified thin section, bone volume, eroded surface, osteoid surface and bone formation rate were calculated for cortical bone of the maxilla, and for cancellous bone of the mandible and proximal tibia. In the OVX-LCa group, compared with the Sham-OVX group, decrease of the bone volume and increase of the bone resorption and formation parameters were detected throughout the observation periods. In the OVX-HCa group, compared with the OVX-LCa group, increase of the bone volume and temporarily increased parameters of bone formation at I week after feeding on high calcium diet were observed in the maxilla, but these changes were not observed in the mandible and proximal tibia. Moreover, the bone resorption and formation parameters in the maxilla, mandible and proximal tibia in the OVX-HCa group became equivalent to the Sham-OVX levels with the passage of time.

  • Histomorphometric analysis on bone dynamics in denture supporting tissue under masticatory pressure in rat

    The bone dynamics under denture base has not been clarified sufficiently. The purpose of this study was to evaluate histomorphometrically the bone dynamics in denture supporting tissue with relation to the intensity of the masticatory pressure. The removable denture bases, which exerted four different regulated masticatory pressures (0.0, 1.0, 20.0 or 40.0 kPa), were inserted to the hard palate of the molar region of 22-week-old rats. The palatal tissues were excised from 1 to 12 weeks after denture insertion at intervals of 1 week and embedded in methyl-methacrylate. No bone resorption was observed throughout the experimental period in the 0.0 and 1.0 kPa groups. In the 1.0 kPa group compared with the 0.0 kPa group, bone formation parameters except for bone formation rate significantly increased at 4 weeks, but there was no significant difference between these groups in the other experimental periods. In the 20.0 and 40.0 kPa groups, bone resorption was observed until 3 and 5 weeks, respectively, and the amounts of bone resorption (AoBR) were 45 +/- 25 and 104+/-21 mum, respectively. After bone resorption, bone formation parameters in these two groups showed transiently significant decrease compared with the 0.0 kPa group, but were the same time course as the 0.0 kPa group following this decrease. In conclusion, masticatory pressure caused different time courses of the bone dynamics in denture supporting tissue depending on the initial intensity of the pressure. The AoBR responded to the initial intensity of masticatory pressure. Bone formation following bone resorption did not cause equivalent recovery of the bone surface level to the level observed in the case without bone resorption in the present study.

  • Effects of ovariectomy and/or dietary calcium deficiency on bone dynamics in the rat hard palate, mandible and proximal tibia

    The effects of ovariectomy and/or dietary calcium deficiency on bone dynamics were examined by comparing the histomorphometric changes in these bones. Five groups of rats were studied, (1) unoperated basal controls: (2) sham-operated, fed on a normal-calcium diet; (3) ovariectomized, fed on a normal-calcium diet; (4) sham-operated, fed on a calcium-deficient diet; (5) ovariectomized, fed on 3 calcium-deficient diet. The basal controls were killed at 6 weeks of ago, and the remaining groups were killed at 18 weeks of age. The hard palate. mandible and proximal tibia were processed undemineralized for quantitative bone histomorphometry. Bone volume, eroded surface, osteoid surface and bone-formation ratio were calculated. A significant age-related increase in bone volume and a significant decrease in bone formation were observed in the hard palate and mandible, whereas no significant age-related increase in bone volume could be found in the tibia. In the hard palate, ovariectomy neither inhibited age-related increases in bone volume nor affected bone dynamics, while both the combined ovariectomy and dietary calcium deficiency and dietary calcium deficiency alone led to bone loss and increased bone turnover. In contrast, in the mandible and proximal tibia, ovariectomy alone as well as dietary calcium deficiency led to bone loss and increased bone turnover. Ovariectomy, therefore, produced no significant changes in the hard palate, but affected bone dynamics in the mandible and tibia. However, dietary calcium deficiency induced bone loss and increased bone turnover in the hard palate, mandible and proximal tibia, independently of ovariectomy. (C) 2001 Elsevier Science Ltd.     This Study examined the effect of masticatory pressure transmitted directly to the hard palate mucosa on the final stage of terminal differentiation of keratinizing system of rats with and without streptozotocin-induced diabetes mellitus. In the nondiabetic rats with masticatory pressure, the number of terminal-deoxynucleotidyl-transferase-mediated deoxyuridine-triphospate-biotin nick end labelling (TUNEL) positive cells tended to increase about twice as much as in the nondiabetic rats without pressure with and without denture. A similar tendency of increase was observed in the diabetic rats without pressure. The synergy of the mechanical pressure and diabetic condition for 2 weeks greatly accelerated the DNA fragmentation, showing 8-fold increase in TUNEL positive cells over the normal control, and caused exfoliation of the stratum corneum. A 4-week exposure of diabetics to the masticatory pressure induced laminar splitting in the midst of the spinosum. Some cells in the stratum granulosum exhibited a sign of DNA fragmentation when laminar splitting took place in the vital cell layer. Premature DNA fragmentation may disturb the adhesion between spinosum cells and prevent the maturation of stratum corneum. Increase in Bax protein-like immunoreactivity in these epithelial cells as revealed by immunocytochemistry may underlie the premature DNA fragmentation in the oral masticatory epithelium under pressure in diabetic patients.

    Osteoclastic bone resorption under the denture base has been considered to be influenced by mechanical force exerted through the denture base. However, no scientific evidence has been shown concerning whether osteoclastic bone resorption is a pressure-threshold-regulated phenomenon or merely a proportionally pressure-dependent phenomenon. In this study, a clear statistical criterion as an objective standard for the evaluation of histopathological bone resorption was established, and the relationship between the amount of continuous or intermittent compressive pressure exerted on the rat hard palate and induced osteoclastic bone resorption was studied based on this criterion. Eighty-five male rats of the Wistar strain, 15 weeks of age, were fitted with experimental dentures which were designed to exert a defined amount of continuous or intermittent compressive pressure on the intermolar region of the hard palate. No bone resorption was observed when continuous compressive pressure was << 1.96 kPa or when intermittent compressive pressure was << 9.8 kPa. In contrast, continuous compressive pressure ≧ 6.86 kPa or intermittent compressive pressure ≧ 19.6 kPa caused significant bone resorption in all rats studied. From these results, it was revealed that osteoclastic bone resorption under the denture base was a pressure-threshold-regulated phenomenon, and that the thresholds exist for the induction of bone resorption under the denture base by both continuous and intermittent compressive pressure, respectively. It was also clearly revealed from the results that the threshold for continuous compressive pressure is lower than that for intermittent compressive pressure.

    The purpose of the present study was to make a histometrical evaluation of bone resorptionunder the denture base caused by lack of self-cleaning as a result of coverage by the denture base.The experimental denture base was designed to make contact with the molar region of the palate of therat and to transmit no occlusal force to the tissues under the denture base. The denture base was not removedthroughout the experimental period. This procedure caused lack of self-cleaning, and it also caused continu-ous pressure to palatal tissues under the denture base by the accumulation of exfoliative horny cells, whichinduced bone resorption under the denture base.Five rats of each denture wearing group and control group were sacrificed every week during theobservation period of TO weeks after the insertion of the experimental denture base. Palatal tissues wereprestained with Villanueva bone stain, and then processed for undecalcified resin section for quantitative bonehistomorphometry. These specimens were analyzed using a two-dimensional graphic analyzing system.From the results of this study, it was revealed histometrically that bone resorption of palatal surface ofhard palate bone tissue initiated under the denture base caused by the lack of self-cleaning as a result ofcoverage by the denture base one week after, became most active two weeks after and ceased five weeks afterthe insertion of the experimental denture base. But on the nasal surface of hard palate bone tissue, boneresorption was not observed.

    Histopathological initial changes in denture supporting tissues, in relation to the changes inintensity of occlusal pressure exerted through the experimental denture base, was studied. The denture basewas applied to the palate of the molar region of the rat, and was installed to cover the palatal mucosa withoutany pressure unless occlusal force was exerted. The defined amount of occlusal pressure was transmitted todenture supporting tissue by allowing the denture base to subside the specified amount into palatal mucosa.The intensity of the occlusal pressure measured immediately after the insertion of the denture were 79. 7, 31. 7and 10.7 kPa. The animals were sacrificed at 2, 4, 7, 10 and 14 days after the insertion of the denture.Histopathological changes caused by the occlusal pressure were observed, and the changes in intensity of theocclusal pressure was also measured on every histopathological observation periods.Histopathological changes of the denture supporting tissues showed dependence on the intensity of theocclusal pressure, and also showed high correlation with the subsequent gradual changes of the pressure.Compression of Iamina propria mucosa, shortening of epithelial ridges, osteoclastic bone resorption, compres-sion of epithelial tissue except for epithelial ridges and inflammatory cell infiltration in the lamina propriamucosa were observed with the elapse of time. These tissue changes came out early and became severe byincreasing the occlusal pressure.

Two to three weeks after the insertion of the denture base, number of osteoclasts and Howship'slacunae were observed on the palatal surface of hard palate bone tissue under the denture base. In the sameobservation period, increased bone formation was observed on the nasal surface.2 . Four to six weeks after the insertion of the denture base, increased bone formation was observed onthe palatal surface of hard palate bone tissue under the denture base. Bone formation was observed mainlyin Howship's lacunae, and only slight bone formation was observed in other region.3 . The first one week and from seven to ten weeks after the insertion of the denture base, bone formationwas suppressed on the palatal surface of hard palate bone tissue under the denture base.

    The initial histopathological change in denture supporting tissue caused by coveringwith denture base without mechanical pressure for the palate of a rat was studied.The purpose of this study was to reveal the relationship between the histopathological changes andthree causative factors : non-pressure contact of denture base with palatal tissue, lack of physiologicat stimulation and lack of self-cleaning for palatal tissue.The experimental denture base was designed to contact the palatal tissue of rat witl?out anymechanical pressure and to transmit no occlusal force to the denture supporting tissue. The palataltissue was examined histopathologically 4, 7, TO and 14 days after the insertion of the. denture base.From the results of this study, it was revealed that non-pressure contact of denture base withpalatal tissue and lack of physiological stimulation would not cause any ittflamrnatory change orbone resorption in the denture supporting tissues except for'the transient slight proliferative reactionin the epithelial ridges. Lack of self-cleaning for palatal tissue, on the contrary, was revealed tocause deformation of some epithelial ridges, decrease of the thickness of Iamina propria mucosae inplaces and bone resorption.

    The histopathological initial changes in the denture supporting tissues were investigated in relation to the impression pressure. The hard palate of adult Wistar rat was employed as experimental area. The experimental area was examined histopathologically and histometrically up to 7 days after the insertion of experimental dentures. Three diverse impression procedures were emploved to make each non-pressure, moderaterressure and heavy-pressure impression. The mean maximum impression pressure for these impressions were 0.0, 9.4 and 22.4g/mm^2, and the mean final impression pressure for these impressions were 0.0, 0.2 and 0.5 g/mm^2, respectively. The pressure which is equivalent to the impression pressure was applied to the denture supporting tissues with the wearing of the denture constructed on the cast poured into each impressions. There was no significant changes during the experimental period in the tissues under the denture which was constructed on the cast poured into non-pressure impression. The histopathological changes in the tissues under the denture which was constructed on the cast poured into moderate or heavy pressure impression came out earlier and more severe with the increase of the pressure. The connective and bone tissues indicated histopathological reaction in advance of the changes observed in the epithelial layer except for epithelial ridges.

    The purpose of this study is to evaluate the usefulness of collagen sponge whose surface layer is reinforced with photopolymerized divinyl adipate (DVA) containing BMP2 as a resorbable bone substitute. No cytotoxicity was observed in the photopolymerized DVA. As a result of implanting the sample on the rat skull, new bone formation was observed in the sample, and significantly more new bone was found in the BMP2-containing sample than in the non-containing sample. From these results, it was suggested that collagen sponge surface-treated with BMP2-containing photopolymerizable DVA shows osteoconductivity, biocompatibility and osteoinductivity, and may be useful as a bone substitute and BMP2 carrier.

    The purpose of the present study was (1) to investigate the changes in spatial learning and memory by tooth loss and occlusal rehabilitation with denture prosthesis, (2) to reveal the effectiveness of theory of spaced learning to the memory. Male Wistar rats were trained on working memory task for 20 days on 8-arm radial maze. After habitation trial, the massed learning group received 8 trials every 5 days. On the contrary, the trial was received twice every 5, 10, 15, and 20 days in the spaced learning group. All groups received the last trial to evaluate the loss of a spatial memory 20 days after these trials. The total number of errors and the time taken to finish the trial were analyzed by ANOVA. Following completion of these behavioral tests, histological investigation was carried out by pyramidal cell count of tissue samples (Nissl staining).
    The massed learning showed significant increase of the number of errors between the eighth and ninth task, but spaced learning didn't show the decrease of the memory. In the ninth task of massed learning groups, hypomnesia progressed in toothless group compared to the other groups. The time was showing a tendency to rise as time passed. The number of hippocampus pyramidal cells in CA1 and CA4 region decreased in order of control, occlusal rehabilitation and toothless groups.In conclusion, tooth loss progressed the decline of the spatial memory, and the occlusal rehabilitation recovered the memory. And it was indicated that the spaced learning would elude the loss of spatial memory compared with massed learning.

    Bisphosphonate(BP)s are high potent inhibitors of osteoclastic bone resorption. We examined whether topical administration of BP could prevent osteoclastic bone resorption under denture base.
    Two hundred and eighty Wistar rats were divided into 3 denture wearing groups (BP(+),BP(-) and covered group) and 1 control group. The experimental denture was installed to cover and to contact with the palatal tissue without any mechanical pressure. In the BP(+) and BP(-) groups, the denture base was designed to exert the masticatory pressure of 20 kPa to the palatal tissue in mastication. In the BP(+) group, 5 μg of BP was injected into the denture supporting tissue for 2weeks before denture insertion. BP(-) group was injected into saline solution. For control group, intentional treatment was not received throughout the experimental period. Fluorescent bone labeling was injected for bone histomorphology. The palatal tissues were excised from 1 to 10 weeks after denture insertion at intervals of 1 week and embedded in methyl-methacrylate. The sections, ground frontally at about 50 μm-thickness at first and second molar region of each palatal tissue, were analyzed histomorphometrically using a two-dimensional graphic-analysis system.
    In the BP(-) and BP(+) groups, eroded surface was larger than the covered group from 1 to 4 weeks and 2 week, respectively. Eroded surface in the BP(+)group was significantly smaller than that in the BP(-) group from 1 to 3 weeks. Amount of bone resorption in the BP(-) and BP(+) groups were 51.8 and 10.7 μm, respectively, and there was a significant difference between these two groups. Therefore, topical administration of BP decreased bone resorption under denture base, and was effective for preservation of residual alveolar ridge.

    In this study, the experimental denture base on the hard palate of the molar region of osteoporotic rats was designed to cover without any mechanical pressure or to load continuous pressure (0.5, 1.5 or 3.4 kPa). Experimental osteoporosis was induced by ovariectomy and feeding on a low calcium diet. The histopathological changes and the changes in the argyrophilic nucleolar organizer region (AgNOR) counts in mucosal epithelium that was revealed to the cellular activity, induced by non-pressure covering or continuous pressure in the tissues uncler the experimental denture bases were examined. Regarding the histological changes induced by continuous pressure, this was evaluated in relation to the time course of the intensity of the continuous pressure. In the tissues under the denture base in osteoporotic condition with non-pressure covering, no histopathological changes caused by nonpressure covering were observed except for a transient slight proliferative reaction in the epithelial ridge. The following histopathological changes were observed in the tissues under the denture base in the osteoporotic condition bearing continuous pressure ; (1) bone resorption was induced on the periosteal surface in the osteoporotic-3.4kPa group, and on the pressure side of the endocortical surface in the osteoporotic-1.5 and -3.4kPa group. (2) histopathological changes in the tissues under the denture base in experimental osteoporosis bearing continuous pressure had an intimate relationship with the initial intensity and the time course of continuous pressure, and with the AgNOR counts in mucosal epithelium.

The purpose of this study was to investigate the effect of diabetes mellitus on bone resorption in the tissues under denture base bearing masticatory pressure by using bone histomorphometry.
    Two-hundred and fifty streptozotocin-induced diabetic male rats (n=50X5 groups) of Wistar strain were used in this study. Denture base was applied to the molar region of the hard palate at 22 weeks of age. In loaded groups, the denture base were designed to load a masticatory pressure of 1.0, 10.0 or 20.0 kPa to the palate, respectively. In covering group, the denture base was designed to contact with the palate without any mechanical pressure. The non-denture-wearing group received no intentional treatment. The cleansing of the palate and removable denture base was carried out at 3 or 4 day intervals. Palatal tissue block was excised from 5 rats of each experimental group after the denture insertion at one week interval. The specimens were stained with Villanueva bone stain, embedded in methyl-methacrylate, and ground frontally to 50μm thickness. Eroded surface and osteoclast surface were calculated by using two-dimensional graphic analyzing system. Amount of bone resorption was calculated as (A)-(B) ; (A) Formed bone tissue during 12 weeks prior to the denture insertion in covering group, (B) Remained bone tissue after the end of bone resorption in each bone resorbed group at each experimental period.
    Bone resorption was observed in 10.0 and 20.0 kPa groups until 6 weeks after the denture insertion, respectively. Total amount of bone resorption was 189±38μm in 10.0 kPa group and 266±27μm in 20.0 kPa group. Comparing these results with results in normal rats in same condition as this study, it is showed that diabetes mellitus prolongs the period of bone resorption and increases total amount of bone resorption.

Amount of bone resorption was calculated as (A) - (B) ; (A) Formed bone tissue during 12 weeks prior to the denture insertion in covering group, (B) Remained bone tissue after the end of bone resorption in each bone resorbed group at each experimental period.
    Bone resorption was observed in 10.0 and 20.0 kPa groups until 3 and 4 weeks after the denture insertion, respectively. Total amount of bone resorption was 60*16mum in 10.0 kPa group and 87*17mum in 20.0 kPa group. Comparing these results with results in normal rats in same condition as this study, it was showed that diabetes mellitus prolongs the term of bone resorption and increases total amount of bone resorption.

    義歯床下組織の変化に関する一連の検討の対象として,我々が選択しているラットの臼歯部口蓋において,骨粗殺症の誘発後に摂取する食餌中のCa含有量と骨動態の変化との関係は明らかにされていない.本研究は,骨粗髭症における義歯床下骨組織の変化に関する検討のための基礎的情報を得ることを目的として,骨粗髭症誘発ラットを対象として,食餌中のCa含有量が臼歯部口蓋骨組織の動態に対して与える影響について組織計測的に検討した.
    ウイスター系雌性ラット108匹(1群27匹×4群)のうちの3群の動物には,6週齢時に卵巣摘出を施すとともに術後12週まで低Ca食(Ca:0.02%)を与えて骨粗穀症を誘発し,術後12週以降は,低Ca食,通常食(Ca:1.15%)および高Ca食(Ca:2.30%)を1群ずつの動物に与えた(低Ca食群,通常食群および高Ca食群).残る1群は対照群として,6週齢時に卵巣摘出の偽手術を施し,その後は通常食を与えた.観察期間は術後12,13,14,16,18,20,22,24および32週とし,各観察期間終了の10日および3日前に骨組織の蛍光ラベリングを施した.各観察期間毎に各群の3匹ずつから採取した臼歯部口蓋組織は,Villanuevaの骨染色を施して未脱灰研磨標本とした.組織計測によって骨量,吸収面,類骨面および骨形成速度のパラメータを求めた.
    低Ca食群では,対照群に比べて観察期間を通じて骨量は有意に低値を示し,その他のパラメータは有意に高値を示した.通常食群ならびに高Ca食群では,術後13〜15週には類骨面ならびに骨形成速度は低Ca食群に比べて同等または有意に大きい値を示したが,吸収面は経時的に減少して,骨量の増加が認められた.また,術後16週以降には類骨面ならびに骨形成速度が減少して,術後18週以降はいずれのパラメータも対照群とほぼ同様の値を示した.これらの結果から,臼歯部口蓋骨組織の骨粗燃症状態を維持するためには,低Ca食飼育が必要であることが示唆された.

    本研究は,骨粗鬆症における義歯床下組織の変化に関する研究の一環として,低カルシウム(Ca)食飼育と卵巣摘出とによって誘発した骨粗鬆症ラットの下顎骨骨組織および臼歯部口蓋骨組織の吸収ならびに形成動態に対して,高Ca食が及ぼす影響について組織計測的ならびに病理組織学的手法を用いて検討した.
    6週齢のウイスター系雌性ラット45匹×2群=90匹に両側の卵巣摘出を施し,施術12週後まで低Ca食(Ca:0.02%)飼育を行い,実験的に骨粗鬆症を発症させた.その後,低Ca食群には施術32週後に至るまで低Ca食を与え続け,高Ca食群には施術12週経過の後は高Ca食(Ca:2.30%)を与えた.観察期間は施術12,13,14,16,18,20,22,24および32週後とし,各観察期間終了の10日前と3日前に骨組織の蛍光ラベリングを行った.各観察期間毎に各実験群の5匹ずつを屠殺して,下顎骨および臼歯部口蓋骨組織を採取し,Villanuevaの骨染色を施した後,前頭断末脱灰研磨標本を作成した.二次元画像解析装置を用いて測定した第一次パラメータに基づいて,第二次パラメータとして分画吸収画率,分画形成面率ならびに石灰化速度を算出した.なお,下顎骨では海綿骨を計測の対象とした.得られた第二次パラメータについて実験群毎に平均値と標準偏差を求め,各観察期間毎に各実験群間の有意差検定(t検定)を行った.
    その結果,骨組織の形成と吸収は,下顎骨では観察期間を通じていずれも経時的に減少しつつほぼ平衡状態を保った.一方,臼歯部口蓋骨組織では,術後13〜16週(高Ca食飼育後1〜4週)には形成量が吸収量を上回り,術後18週以降にはほぼ平衡状態を示した.本研究の結果は,本研究の同一条件下で施術12週後以降に観察された下顎骨における骨量の維持ならびに臼歯部口蓋骨組織における骨量の増加(原ら,1996)と整合性を示すものである.

    本研究は,ストレプトゾトシン(STZ)誘発糖尿病ラットの臼歯部口蓋を対象として,義歯床を介して加えられる咀嚼圧によって義歯床下組織に惹起される病理組織学的変化に対して糖尿病が与える影響について,組織計測的ならびに病理組織学的手法を用いて検討するとともに,咀嚼圧の大きさとの関連において検討を加えることを目的とした.
    STZによって糖尿病を誘発した15週齢のウイスター系雄性ラット155匹を用いた.安静時には義歯床下組織と無圧の状態で接触し,咀嚼時には100,50または13μm沈下して義歯床下組織に咀嚼圧を加える義歯床を1群(40匹)ずつの動物に装着した.義歯床および義歯床下粘膜の清掃は3〜4日毎に行った.対照群(35匹)のラットについては無処置のまま経過させた.義歯床装着の3日,1,2,4,8,12および20週後に各群の5匹ずつを屠殺して義歯床下組織を採取し,通法に従って第1臼歯部において前頭的な4μmのパラフィン切片を作成し,これにヘマトキシリン-エオジン染色を施して光学顕微鏡下で病理組織学的変化を観察した.組織計測によって上皮組織の面積,粘膜固有層の面積および破骨細胞数を求めた.また,義歯床下組織に加えられる咀嚼圧の大きさは,各義歯装着群の残る5匹ずつを対象として,義歯床装着時,3日後および1週後以降1週毎に20週後まで測定した.得られた結果を,健常ラットを対象とした同一条件下の研究結果(原,1991)およびSTZ誘発糖尿病ラット臼歯部口蓋の組織変化(白井,1994)と比較しつつ検討を加えた.
    その結果,糖尿病は,咀嚼圧によって義歯床下組織に惹起される病理組織学的変化に対して次の影響を与えた.1)上皮細胞間の結合を脆弱化し,その程度を経時的に増強した.2)上皮組織の退行性ならびに増殖性の変化を減弱した.3)粘膜固有層の圧扁を増強した.4)炎症性細胞浸潤を増強した.5)炎症性細胞浸潤を惹起する咀嚼圧の閾値を低下した.6)破骨細胞の出現数を増加し,その発現期間を延長した.6)破骨細胞の出現をもたらす咀嚼圧の閾値を低下した.8)骨芽細胞の出現を阻害した.9)咀嚼圧の経時的減少率を増加した.本研究の結果から,糖尿病は義歯床を介して加えられる咀嚼圧に対する義歯床下組織の耐性を低下させることが示唆された.

    本研究はラットの臼歯部口蓋を対象として,義歯床を介して加えられる咀嚼圧によって惹起される義歯床下粘膜上皮細胞の活性動態の変化について,上皮基底細胞核が示すNORs(nucleolar organizer regions)数を指標として,上皮組織に惹起される病理組織学的変化との関連において検討することを目的とした.
    15週齢のウイスター系雄性ラット120匹(義歯装着群3群と対照群1群 各30匹)を用いた.義歯装着群に対しては,安静時には義歯床下組織と無圧の状態で接触し,咬合時には規定量(100,50および13μm)だけで沈下する義歯床を装着した.対照群は義歯を装着することなく経過させた.義歯床装着1,2,4,8,12および20週後に各群の5匹ずつを屠殺して義歯床下の口蓋粘膜を採取し,4%パラホルムアルデヒドを用いて固定した後,通法に従ってパラフィン包埋した.2μmのAgNOR染色標本を対象として,各実験条件毎に上皮基底細胞核1個当たりのNORs数の平均値を求め,各実験群間の有意差検定を行った.また,4μmのヘマトキシリン-エオジン染色標本を対象として上皮組織の病理組織学的変化を観察した.
    その結果,義歯床下粘膜の上皮基底細胞核の示したNORs数は,病理組織学的変化との関連を示して変化した.義歯床下粘膜上皮に退行性の組織変化がみられた義歯床装着1および2週後のNORs数は,いずれの義歯装着群においても対照群に比べて有意に減少すると同時に,義歯床の沈下量の増加に伴って減少した.4週後の義歯装着群のNORs数はいずれも,対照群に比べれば有意に低い値を示すものの,上皮組織の増殖性変化に伴って2週後に比べて増加して各沈下群ともほぼ同様の値を示した.8週後以降は,13μm沈下群のNORs数が経時的にわずかに増加して対照群と同様の値を示したが,50および100μm沈下群では対照群ならびに13μm沈下群に比べて,その差は大きくはないものの,有意に低い値を示した.

    本研究は,卵巣摘出と低Ca食摂取によって実験的に誘発した骨粗鬆症ラットを対象として,高Ca食を摂取させた場合の骨量変化を組織計測的手法を用いて比較検討し,実験的骨粗鬆症の顎骨における骨量変化に対して高Ca食が及ぼす影響を明らかにすることを目的とした.
    ウイスター系雌性ラット54匹(低Ca食群ならびに高Ca食群各27匹)を用い,6週齢時に両側の卵巣を摘出した.施術12週後までは各実験群とも低Ca食(Ca:0.02%)を与え,その後低Ca食群には施術に32週後至るまで低Ca食を与え続け,高Ca食群には施術12週後以降は高Ca食(Ca:2.3%〜通常食の2倍)を与えた.飲料水は各群とも蒸留水を与えた.施術12,13,14,16,18,20,22,24,および32週後に各群の3匹ずつを屠殺して,硬口蓋骨組織(上顎骨と口蓋骨),下顎骨ならびに脛骨近心端を採取し,Villanuevaの骨染色を施した後にMMA樹脂に包埋し,約50μmの非脱灰研磨標本とした.各実験例毎に全骨組織量(=全骨量+全骨髄量)を測定し,単位骨量=全骨量/全骨組織量を算出した.各実験群毎に全骨量と単位骨量の平均値と標準偏差を求め,各実験群間の有意差検定を行った.
    その結果,下顎骨ならびに脛骨近心端の海綿骨では観察期間を通じて高Ca食群における骨量の増加はみられなかった.しかし,硬口蓋骨組織では高Ca食群における骨量は,施術18週後以降増加の傾向を示し,20週後以降は低Ca食群に比べて有意に増加した.したがって,卵巣摘出と低Ca食とによって誘発した骨粗鬆症の硬口蓋骨組織における骨量の増加に対しては,高Ca食の摂取が効果を示すことが示された.

    その結果,骨粗鬆症群では対照群に比べて,両観察期間を通じて骨量の明らかな減少がみられ,骨表面には破骨細胞による骨吸収と骨芽細胞による骨添加が生じている部位が多く認められた.単位骨量は対照群に比べて減少し,分画形成面率,分画吸収面率および石灰化速度は対照群に比べて増加した.
    以上の結果から,本研究で得られた骨粗鬆症の硬口蓋骨組織は,骨吸収と骨添加を増加させる高代謝回転型の変化を示すことが明らかになった.

    6週齢のウイスター系雌性ラット30匹を各群10匹ずつの卵巣摘出(OVX)群、低カルシウム(低Ca)食摂取群および対照群に分けた。OVX群は両側の卵巣を摘出し、他の2群に対しては偽手術を行った。低Ca群には低Ca食(Ca:0.02%)と蒸留水を、他の2群には通常食(Ca:1.15%)と蒸留水を与えた。観察期間は術後12および32週(18および38週齢)とし、屠殺の10日前にテトラサイクリン(体重100gあたり5.0mg)を、3日前にカルセイン(体重100gあたり0.8mg)を投与した。各観察期間毎に各実験群の5匹ずつを屠殺して硬口蓋骨組織を採取し、Villanuevaの骨染色を施した後、MMA樹脂に包埋し、頬舌的な厚さ50μmの未脱灰研磨標本を作製した。落射蛍光顕微鏡と二次元画像解析装置を用いて単位骨量、分画形成面率、分画吸収面率および石灰化速度などのパラメーターを求め、低Ca食およびOVXによって誘発された骨粗鬆症が硬口蓋骨組織の動態に与える影響について検討を加えた。
    その結果、OVX群においては両観察期間とも、いずれのパラメーターについても対照群との間に差はみられなかった。低Ca食摂取群においては両観察期間を通じて、単位骨量は対照群およびOVX群に比べて減少し、分画形成面率、分画吸収面率および石灰化速度は対照群およびに比べて増加した。
    以上の結果から、硬口蓋骨組織における骨粗鬆症状態の誘発にはOVXのみでは十分な効果は得られず、低Ca食の摂取を必要とした。また、本研究で得られた骨粗鬆症の硬口蓋骨組織は、骨吸収と骨添加の増加による高代謝回転型の変化を示すことが明らかになった。
